摘 要:【目的】探讨栀子苷对缺氧/复氧心肌细胞前炎症因子肿瘤坏死因子-α(TNF-α)、白细胞介素-1(IL-1)、IL-6释放以及对Toll样受体4(TLR4)和活化核因子-κB(NF-κB)蛋白表达的影响。【方法】原代培养SD大鼠乳鼠心肌细胞。实验分为正常对照组、缺氧/复氧模型组和栀子苷预处理组,采用酶联免疫吸附(ELISA)法检测细胞培养液前炎症因子TNF-α、IL-1、IL-6的含量,Western blot法检测心肌细胞TLR4、NF-κB蛋白表达。【结果】与缺氧/复氧模型组比较,栀子苷预处理组心肌细胞TNF-α、IL-1、IL-6释放显著减少,TLR4、NF-κB蛋白表达显著下降,差异均有统计学意义(P<0.05或P<0.01)。【结论】栀子苷可抑制缺氧/复氧心肌细胞前炎症因子TNF-α、IL-1、IL-6释放,其作用机制可能与栀子苷下调TLR4-NF-κB通路有关。Objective To explore the effect of geniposide on the release of pro-inflammatory cytokines tumor necrosis factor-alpha(TNF-α),interleukin(IL)-1 and IL-6 in hypoxia/reoxygenation myocardial cells and on the expression of Toll- like receptor(TLR) 4 and nuclear factor-kappa B(NF-κB) of the myocardial cells.Methods Primary rat cardiac myocytes were cultured. And the cells were randomly divided into 3 groups,namely normal control group, hypoxia/reoxygenation group and geniposide-pretreatment group. The contents of TNF-α,IL-1,and IL-6 were determined by enzyme-linked immunosorbent assay(ELISA),and the protein expression levels of TLR4 and NF-κB were detected by Western blotting method. Results Compared with the hypoxia/reoxygenation group,the geniposide-pretreatment group reduced the release of TNF-α, IL-1, IL-6(P〈0.05) and decreased the expression of TLR4 and NF-κB(P〈0.05 or P〈0.01). Conclusion Geniposide can inhibit the release of pro-inflammatory cytokines in the anoxia/reoxygenation myocardial cells, and its mechanism may be associated with the down-regulation of TLR4-NF-κB signaling pathway.
