摘  要:目的:研究复肾功方对慢性肾功能衰竭(chronic renal failure,CRF)大鼠肾组织α-平滑肌肌动蛋白(α-SMA)表达的影响,探讨其减轻肾间质纤维化的作用机制。方法:将55只雄性SD大鼠随机分为正常组,模型组,复肾功方低、中、高剂量组,每组11只。正常组常规饲养,其余四组大鼠食用含0.5%腺嘌呤饲料制造慢性肾功能衰竭模型,连续造模21d。造模成功后,所有大鼠改用常规饲料。正常组和模型组按20 m L/kg灌胃生理盐水,复肾功方低、中、高剂量组分别以4、8、16 g生药/kg体质量灌胃复肾功方,1日1次,连续30d。实验结束后,检测各组大鼠血清血肌酐(Scr)和尿素氮(BUN)水平,Masson染色检测肾间质纤维化程度,免疫组化和Western blot法检测α-SMA蛋白的表达。结果:与正常组比较,模型组大鼠Scr和BUN水平及肾组织α-SMA表达明显升高(P<0.05),肾间质出现明显纤维化病变。经复肾功方治疗后,各剂量组大鼠Scr和BUN水平及肾组织α-SMA表达明显降低。结论:复肾功方能显著改善CRF大鼠肾纤维化病变,其机制可能与降低肾组织α-SMA表达有关。AIM: To investigate the effect of Fushengong Decoction on the expression of α-SMA in renal tissue of rats with chronic renal failure( CRF),to study the mechanism of reducing renal interstitial fibrosis.METHODS: 55 male SD rats were randomly divided into five groups: control group,model group,and low,medium and high Fushengong Decoction dose groups(11 in each group).Rats in control group were fed with standard chow,while the other four groups were fed with 0.5% adenine feed stuff to make the CRF model.After the models were successfully made,all rats received standard chow.Then,rats in control and model groups received intra-gastric normal saline( NS) by 20 mL·kg^-1·d^-1 for 30 d,while those in low,medium and high Fushengong Decoction dose groups received Fushengong Decoction at the dose of 4,8 and 16 g/kg respectively,once a day for 30 d.After the experiment,blood urea nitrogen( BUN) and serum creatinine( Scr) of rats were detected in each group.The renal interstitial fibrosis was detected by Masson staining,and the expression of α-SMA was detected by immunohistochemistry and Western blot method.RESULTS: The levels of BUN and Scr and the expression of α-SMA in nephridial tissue increased significantly in the model group compared to those of the control group( P 〈0.05),and the renal interstitium showed the fibrotic lesions in model group.After the treatment of Fushengong decoction,comparing with the model group,the levels of BUN and Scr and the expression of α-SMA decreased significantly( P〈0.05).CONCLUSION: Fushengong Decoction can significantly improve the renal fibrosis in rats with CRF by reducing the expression of α-SMA in nephridial tissue.
