摘  要:目的:研究羟基红花黄色素A(HSYA)对β淀粉样蛋白(Aβ)诱导的氧化应激和学习记忆功能损伤的影响。方法:通过侧脑室注射Aβ建立学习记忆功能损伤的动物模型,术后通过腹腔注射HSYA进行干预,采用Morris水迷宫实验来检测学习记忆功能的改变,测定大鼠脑组织中丙二醛(MDA)、超氧化物歧化酶(SOD)和还原型谷胱甘肽(GSH)的含量以及炎症因子IL-6和TNF-α的水平。结果:HSYA能够改善由Aβ诱导的学习记忆功能损伤,降低脑组织海马中MDA的含量,增加SOD和GSH的含量;降低脑组织海马中IL-6和TNF-α的水平。结论:HSYA能抑制由Aβ诱导的脑组织中氧化应激水平和炎症反应,改善由Aβ诱导的学习记忆功能损伤。AIM: To study the effects of hydroxysafflor yellow A( HSYA) on β-amyloid-induced oxidative stress and dysfunction of learning and memory.METHODS: Rats were randomly divided into four groups: control group,sham operation group,NS( normal saline) group and HSYA group.The animal model for dysfunction of learning and memory was established by intracerebroventricular injection of β-amyloid in NS and HSYA groups.The HSYA group was received 6 mg/kg per day HSYA intraperitoneally for 14 days after intracerebroventricular injection.The other three groups were received the same amount normal saline for 14 days after operation.The Morris water maze was used to evaluate the function of spacial learning and memory.The levels of MDA,SOD,GSH,IL-6 and TNFin rats' hippocampus were measured after the Morris water maze test.RESULTS: HSYA reversed β-amyloid-induced cognitive impairment.HSYA reduced the levels of MDA,IL-6,TNF-α and increased the levels of SOD,GSH in rats ' hippocampus.CONCLUSION: HSYA can attenuate β-amyloid-induced oxidative stress reaction and reverse β-amyloid-induced cognitive impairment in brain of rats.
