β-酪啡肽7对链脲佐菌素诱导糖尿病肾病模型大鼠的肾保护

摘　　要：背景:研究发现β-酪啡肽7有对糖尿病模型大鼠减轻肾小管上皮细胞的氧化及降血糖的作用,但关于β-酪啡肽7对糖尿病肾病作用机制的研究比较少。目的:探讨β-酪啡肽7对糖尿病肾病模型大鼠肾损伤的作用。方法:将30只大鼠随机以n=10等分为对照组、模型组和β-酪啡肽7组。构建糖尿病模型大鼠20只,造模成功后模型组和β-酪啡肽7组模型大鼠腹腔注射链脲佐菌素(60 mg/kg),对照组腹腔注射等量柠檬酸液,β-酪啡肽7组模型大鼠给予β-酪啡肽7灌胃30 d,模型组和对照组给予等量生理盐水灌胃。结果与结论:1与对照组相比,模型组大鼠胰岛素水平降低,血糖、尿糖、尿蛋白、血清肌酐、血清尿素氮水平以及肾脏中Ⅰ型胶原蛋白和Ⅳ型胶原蛋白的mRNA和蛋白表达水平增加。2与模型组相比,β-酪啡肽7组模型大鼠上述指标明显恢复。表明β-酪啡肽7对糖尿病肾病模型大鼠肾损伤有保护作用。BACKGROUND： Studies have found that β-casomorphin 7 can lessen oxidation of renal tubular epithelial cells and lower blood sugar in a rat model of diabetes. There is a few studies concerning the effect of β-casomorphin-7 on diabetic nephropathy. OBJECTIVE： To explore the effects of β-casomorphin-7 on renal injury in a rat model of diabetic nephropathy. METHODS： The 30 rats were equally and randomly divided into control group, model group and β-casomorphin-7 group. Twenty rat models of diabetes were established. In the model and β-casomorphin-7 groups, rats were intraperitoneally injected with streptozotocin（60 mg/kg）. In the control group, rats were intraperitoneally injectedwith an equal volume of citric acid. Rats in the β-casomorphin-7 group were intragastrically administered β-casomorphin-7 for 30 days. Rats in the model and control groups were given an equal volume of saline by intragastric administration. RESULTS AND CONCLUSION：（1） Compared with the control group, insulin levels decreased, glucagon, blood glucose, urine glucose, urine protein, serum creatinine, serum urea nitrogen levels, m RNA and protein expression levels of type I collagen and type IV collagen in kidney increased in the model group.（2） Compared with the model group, above indexes were apparently recovered in the β-casomorphin-7 group. These findings indicate that β-casomorphin-7 has a protective effect on renal injury in diabetic nephropathy rats.
