大鼠血浆艾芬地尔HPLC-MS测定方法的建立

摘　　要：通过采用C18色谱柱（5μm,4.6 mm×150 mm）,以甲醇-乙酸铵溶液（p H值7.20）为流动相,梯度洗脱（0-3 min,20%A→90%A,80%B→10%B）,流速0.6 ml/min,以酮康唑为内标。质谱检测（MS）设定正离子模式（Positive）,采用电喷雾离子源（ESI）,选择离子检测方式（SIM）,艾芬地尔和内标的最佳目标检测离子分别为m/z 326.3、m/z 531.0。建立了高灵敏度的HPLC-MS法检测大鼠血浆中艾芬地尔。Ketoconazole is used as internal standard. The C 18 column（ 5μm,4. 6 mm × 150 mm） is used with the mobile phase of methanol-ammonium acetate solution（ p H 7. 0） and gradient elution（ 0- 3 min,20% A→90% A,80% B→10% B） at the flow rate of 0. 6 ml / min. The detection is performed in single ion monitor（ SIM） in positive ionization mode with an electrospray ionization（ ESI） source. The single ion monitored is respectively m / z 326. 3 for ifenprodil and m / z 531. 0 for ketoconazole. An HPLC-MS method for the determination of ifenprodil in rat plasma is established.
