摘  要:目的研究基于毛细管电泳的鹿鞭DNA指纹鉴定方法。方法盐析法提取梅花鹿鞭、马鹿鞭、驯鹿鞭和赤鹿鞭的线粒体DNA,对序列为GAGCGTCGAA的引物进行RAPD扩增,毛细管电泳法分析其产物,再对所得指纹图谱的相似度进行分析。结果梅花鹿鞭指纹图谱中有9个共有峰,马鹿鞭指纹图谱中有21个共有峰,与驯鹿鞭和赤鹿鞭有明显差异。而且,其相似度均小于0.7。结论该方法客观、准确、方便,可有效区分梅花鹿鞭、马鹿鞭及其伪品(驯鹿鞭、赤鹿鞭)。AIM To study the DNA fingerprint identification of Penis et Testis Cervi based on capillary electrophoresis. METHODS The mitochondrial DNAs of Penis et Testis Cervi of Cervus nippon Temminck,Cervus elaphus Linnaeus, Rangifer tarandus and Cervus canadensis were extracted by salting-out method. The primer( GAGCGTCGAA) was used for RAPD amplification,whose products were analyzed by capillary electrophoresis.Then the similarities of obtained fingerprints were analyzed. RESULTS There were nine and twenty-one common peaks in the fingerprints of Penis et Testis Cervi of C. nippon and C. elaphus,respectively,having obvious difference from those of R. tarandus and C. canadensis. In addition,their similarities were less than 0. 7. CONCLUSION This method is objective,accurate and simple,which can effectively distinguish C. nippon and C. elaphus from their counterfeits( R. tarandus and C. canadensis).
