摘  要:【目的】建立利用SCAR标记技术鉴定番茄花叶病毒病抗性基因Tm-2^2的技术体系,开展分子标记辅助选择育种,选育加工番茄抗病品种。【方法】以6份To MV表现型鉴定已知的加工番茄材料为试材,选用与Tm-2^2基因连锁的SCAR标记,通过PCR扩增和Hind III酶切鉴定有无Tm-2^2基因,并将其应用于加工番茄品种选育。【结果】抗感试材均产生950 bp和1 100 bp的特异片段,纯合抗病基因型无Hind III酶切位点,杂合抗病基因型和感病基因型有Hind III酶切位点,酶切结果为:纯合抗病1 100 bp+950 bp、杂合抗病1 100 bp+950 bp+500 bp+300 bp+150 bp、感病基因1 100 bp+500 bp+300 bp+150 bp。【结论】通过酶切产生的特异性片段就可鉴定出Tm-2^2基因,成本低,操作不受时间、地域、发育时期的限制,提高了选择的准确性,加快了育种进程。【Objective】This research attempts to identify Tm- 2^2 gene which is resistant to tomato mosaic virus by SCAR markers and a technical system for molecular marker- assisted breeding is intended to be established in the hope of providing a theoretical basis for disease resistant varieties breeding in Solanum lycopersicum.【Method】The SCAR markers which is tightly linked with Tm- 2^2 gene were used to amplify genome DNAs of six materials( the result of phenotype identification has been already known),using PCR amplification and Hind III enzyme to identify whether it contains Tm- 2^2 gene or not. After identification,the resistant breeding took advantage of it. 【Result】Both resistant and susceptible materials produced 950 bp and 1,100 bp fragment. The amplified bands from susceptible and heterozygous were distinguishable after cleavage with the restriction enzyme Hind III. Genotype susceptible and heterozygous with Tm- 2^2 gene could produce respectively 1,100,500,300 and 150 bp and 1,100,950,500,300 and 150 bp,homozygous genotypes still presented 1,100 and 950 bp fragment.【Conclusion】According to the products which was digested by HindⅢ enzyme we can make sure whether it contains Tm- 2^2 gene or not. The result of this method is obvious and reliable with low cost,not limited by time,region or growing periods,and it improves the accuracy of the selection and speeds up the breeding process.
