SCID与NOD/SCID小鼠部分免疫指标的比较

摘　　要：目的测定SCID和NOD/SCID小鼠的免疫器官质量、血常规指标以及部分血液生化指标（immunoglobulin G,IgG）,分析比较2个免疫缺陷小鼠在免疫方面的特点。方法对4~8、12~16和20~24周龄SCID与NOD/SCID小鼠眼眶静脉采血,测定血常规指标,剖取胸腺和脾脏进行称质量比较。对4~6、8~10、16~18周龄的SCID和NOD/SCID小鼠眼眶静脉采血,测定比较血清IgG含量。结果 a.同周龄SCID与NOD/SCID小鼠胸腺质量差异无显著性。3个年龄段（4~8、12~16和20~24周龄）的NOD/SCID小鼠脾脏显著低于各组SCID小鼠（P〈0.05）,SCID小鼠脾脏随年龄增加趋势明显;b.NOD/SCID小鼠白细胞数（white blood cell count,WBC）、淋巴细胞绝对值（lymphocytes,LYM#）、红细胞数（red blood cell,RBC）、血红蛋白含量（hemoglobin,HGB）及平均血红蛋白浓度（mean corpuscular hemoglobin concentration,MCHC）显著低于同周龄SCID小鼠（P〈0.05）,高龄SCID小鼠白细胞水平和淋巴细胞百分比显著高于低龄（4~8周龄）SCID鼠（P〈0.05）。c.NOD/SCID小鼠血清IgG含量显著低于SCID小鼠（P〈0.05）。结论SCID与NOD/SCID小鼠部分免疫指标存在显著性差异。Objective To analyze and compare some immunological characteristics by determining the weight of immune organs,blood indexes and some blood biochemical parameters（IgG）of SCID and NOD/SCID mice.Methods The thymus and spleen of 4-8,12-16 and 20-24 week-age SCID and NOD/SCID mice were dissected to compare their organ weights,and their orbital venous blood samples were taken for blood routine examination.In addition,the orbital venous blood of 4-6,8-10 and 16-18 week-age SCID and NOD/SCID mice were taken for serum IgG determination.Results a.There was no significant difference between the thymus weights of SCID and NOD/SCID mice.The spleen weight of NOD/SCID mice at 4-8,12-16 and 20-24 week age was significantly lower than that of SCID mice（P〈0.05）.The spleen weight of SCID mice was increasing along with age.b.Compared with SCID at the same age,WBC,LYM#,RBC,HGB and MCHC in blood sample of NOD/SCID mice were significant lower（P〈0.05）,but WBC and LYM（%）were significantly increased in SCID mice with age.c.NOD/SCID mice showed significantly lower content of serum IgG（P〈0.05）.Conclusions SCID and NOD/SCID mice display significant difference in some immune indexes.
