摘 要:目的:观察益气活血通络方对糖尿病周围神经病变大鼠背根神经节神经生长因子受体p75NTR和TrkA表达的影响,以期探讨益气活血通络方治疗糖尿病周围神经病变的分子机制。方法:选用SD大鼠,采用小剂量链脲佐菌素(35 mg/kg)联合高脂饲料复制糖尿病周围神经病变大鼠模型。将造模成功的大鼠随机分为模型组,益气活血通络方高、中、低剂量治疗组和甲钴胺阳性对照组,另设正常对照组。各组用药后,采用酶联免疫吸附法(ELISA)测定大鼠血清NGF含量,Western blot法检测背根神经节p75NTR和TrkA蛋白的表达。结果:与正常组比较,糖尿病周围神经病变大鼠背根神经节的p75NTR表达水平明显升高(P﹤0.01),血清神经生长因子(NGF)含量、背根神经节的TrkA表达水平明显降低(P﹤0.01)。益气活血通络方能明显降低糖尿病周围神经病变大鼠背根神经节的p75NTR水平(P﹤0.01或P﹤0.05),并升高NGF、TrkA水平(P﹤0.01),各剂量组间无统计学差异。结论:益气活血通络方可通过抑制糖尿病周围神经病变大鼠背根神经节NGF低亲和力受体p75NTR的表达,激活NGF及其高亲和力受体TrkA的表达,促进神经元生长。AIM: To investigate the molecular mechanism of Yiqihuoxuetongluo recipe in the treatment of diabetic peripheral neuropathy( DPN),and also the effect of Yiqihuoxuetongluo recipe on the expression of p75NTR and TrkA in nerve growth factor( NGF) receptor of dorsal root ganglion( DRG) of DPN rats. METHODS: SD rats were randomly divided into 6 groups: the normal control group,the model group, Yiqihuoxuetongluo recipe treatment groups( 9,4. 5,2. 25 g·kg^(-1)·d^(-1),i. g.),and Mecobalamine treatment group( 0. 225mg·kg^(-1)·d^(-1)). DPN rat model was established by given SD rats Streptozotocin( STZ)( 35 mg/kg)and high-fat diet. After the treatment, enzyme linked immunosorbent assay( ELISA) and Western blot were used to detect the content of serum NGF and the expression of p75NTR and TrkA proteins in DRG. RESULTS: Compared with the normal control group,the expression level of p75NTR in DRG of diabetic rats increased significantly( P ﹤0. 01),while serum NGF and the expression of TrkA in DRG decreased significantly in diabetic rats( P ﹤0. 01). Yiqihuoxuetongluo recipe significantly reduced p75NTR levels( P ﹤0. 01 or P ﹤0. 05) and increased serum NGF and TrKA levels( P ﹤0. 01)in DRG of diabetic rats. The effect of Yiqihuoxuetongluo recipe was not dose-dependent. CONCLUSION: The results demonstrated that Yiqihuoxuetongluo recipe can promote the growth of neurons through inhibition of the expression of low affinity receptor of NGF in DRG of diabetic rats,p75NTR and activation of NGF and the expression of its high affinity receptor,TrkA.
