摘  要:目的观察阿托伐他汀对人结肠癌细胞株(LoVo)细胞增殖及凋亡的影响,探讨其机制。方法用不同浓度的阿托伐他汀培养基处理人结肠癌LoVo细胞,分为3组:低剂量组(20p.mol/L)、中剂量组(60μmol/L)、高剂量组(100μmol/L)。采用噻唑蓝(MTY)比色法检测阿托伐他汀对人结肠癌LoVo细胞增殖的抑制率;采用流式细胞仪检测阿托伐他汀对人结肠癌LoVo细胞的凋亡率;采用反转录-聚合酶链反应(RT—PCR)法检测人结肠癌LoVo细胞中B细胞淋巴瘤/白血病-2(bcl-2)mRNA的表达。结果阿托伐他汀能够显著抑制LoVo细胞的增殖,促进其凋亡,并且此作用随着阿托伐他汀浓度的增加而增强[(30.40±1.28)%、(51.48±1.20)%、(69.14±1.42)%;(5.95±0.13)%、(20.38±0.33)%、(29.87±0.32)%],阿托伐他汀还能够下调bcl-2基因的表达。结论阿托伐他汀能够诱导人结肠癌LoVo细胞的凋亡,这可能与其下调bcl-2基因的表达有关。Objective To observe the effect of atorvastatin on proliferation and apoptosis of colon cancer LoVo cells and explore the possible mechanisms. Methods The LoVo cells were treated with the different concentrations of atorvastatin and divided into three groups: the low concentration group (20 μmol/L), the middle concentration group (60 μmol/L) and the high concentration group (100 μmol/L). The growth inhibition rate was calculated by MTT assay. The cell apptosis rate was as- sessed by flow cytometry. The expression of B cell lymphoma/leukemia -2 (bcl -2) was detected by re- verse transcription - polymerase chain reaction ( RT - PCR). Results The atorvastatin could inhibit the growth of the LoVo cells and prompt the LoVo ceils apoptosis in a concentration - dependent manner [(30.40±1.28)%, (51.48±1.20)%, (69.14±1.42)%; (5.95 ±0.13)%, (20.38±0.33)%, ( 29. 87 ± 0. 32 ) % ]. The expression of bcl - 2 was downregulated with the increase of the atorvastatin con- centration. Conclusion Atorvastatin could inhibit the growth of LoVo cells possibly by inducing apoptosis through downregulating bcl -2 mRNA expression.
