不同材质人工血管在犬体内应用的效果对比研究

摘  要:目的分析研究涤纶人工血管、真丝人工血管、膨胀聚四氟乙烯(eFFFE)人工血管及CD34^+细胞种植ePTFE人工血管在犬体内的应用效果。方法取健康杂种家犬共20条,分为4组,分别植入涤纶人工血管、真丝人工血管、膨胀聚四氟乙烯ePTFE人工血管、CD34^+细胞种植ePTFE后人工血管,均选择犬下腔静脉、腹主动脉进行植入。对比术前、术后不同时间血小板计数、前列腺素F1α(PGF1α)、血浆血栓素B2(P/T)比值、通畅率、新生内膜厚度等。结果CD34^+细胞组血小板浓度低、血清PGF1α高,P/T比值高,该组术后1个月的血小板浓度、血清PGF1α、P/T比值分别为(302.17±9.75)/L、(176.28±7.87)pg/L、0.31±0.04,与其他组别比较,P〈0.05,差异有统计学意义。CD34^+细胞组动、静脉血管闭塞率、狭窄率均低于其他组,人工血管内膜厚度小于其他组别,其中CD34^+细胞组的动脉闭塞率与狭窄率分别为0.00%与30.00%;静脉分别为30.00%与50.00%;CD34^+细胞组的动静脉新生内膜厚度分别为(146.27+3.99)μm和(190.09±8.86)μm,分别与其他组别比较差异有统计学意义(P〈0.05)。CD34^+细胞组人工血管腔面内膜内皮细胞排列均匀完整,其他组别欠完整。结论纯化后CD34^+细胞种植ePTFE可以保持较为理想的通畅率和内皮化,有效抑制PGF1α,减少血小板过度升高,避免血栓形成。Objective Analysis of polyester artificial blood vessels, artificial blood vessels silk, expanded polytetrafluoroethylene (ePTFE) grafts and CD34^+ cells were seeded ePTFE application results in dogs. Methods a total of 20 mongrel dogs were divided into 4 groups, namely polyester implanted arti- ficial blood vessels, artificial blood vessels silk, expanded polytetrafluoroethylene ePTFE grafts, CD34^+ cells after planting ePTFE vascular grafts are inferior vena cava select dog abdominal aortic implantation. Compare before surgery, different time platelet count, PGF1α, PiT ratio, the patency rate, neointimal thickness. Results CD34^+ cells group of low platelet concentration, high serum PGF1 alpha, P/T ratio is high, the group after 1 month of platelet concentration, serum PGF1 alpha, the P/T ratio (302. 17 ± 9. 75)/L respectively, (176. 28 ±7. 87) pg/L, 0. 31 ±0.04, compared with other groups, P 〈0.05, the difference was statistically significant. CD34^+ cells group dynamic, and narrow vein occlusion rate were significantly lower than other groups, artificial vascular intima thickness is less than the other group, which the artery occlusion rate of CD34^+ cells group and narrow rate were 0.00% and 30.00% respectively; Veins are 30. 00% and 30. 00% respectively; Arteriovenous neointimal thickness of CD34^+ cells group, re- spectively (146. 27 ± 3.99) μm and (190. 09 ± 8. 86) μm, significant difference compared with other groups data respectively (P 〈 0. 05). CD34^ + ceils group of artificial vascular cavity surface lining endothelial cells arranged evenly complete. Conclusion The purified CD34^+ cells were seeded ePTFE maintain ideal paten- cy rate and endothelialization, inhibit PGF1α, thrombocytopenia excessive rise, avoid thrombosis.
