摘  要:目的探讨谷胱甘肽(GSH)减少大鼠隐睾生精细胞凋亡的机制。方法将健康雄性sD大鼠30只。随机分为3组:对照组、隐睾组、GSH治疗组,采用“3周尾吊”法复制大鼠隐睾模型,采用DNA断裂的原位末端标记(TUNEL)法检测生精细胞凋亡指数(AI),采用化学比色法检测睾丸组织谷胱甘肽过氧化物酶(GSH—Px)活性、总抗氧化能力(T—AOC)及丙二醛(MDA)含量。结果隐睾组中睾丸重量、GSH—Px活性及T—AOC均明显低于对照组[(0.577±0.019)g,(22.68±1.22)U/mgprot,(11.9±1.4)U/mg;(1.804±0.022)g,(31.68±0.85)U/mgprot,(29.7±2.5)U/mg],生精细胞AI、MDA含量均高于对照组[(1.73±0.06)%、(2.72±0.13)nmol/mgprot;(0.55±0.05)%、(0.53±0.07)nmol/mgprot],GSH治疗组中睾丸重量、GSH—lax活性及T-AOC均明显高于隐睾组[(1.285±0.012)g、(32.83±0.95)U/mgprot、(30.9±1.8)U/mg;(0.577±0.019)g、(22.68±1.22)U/ragprot、(11.9±1.4)U/mg],生精细胞AI、MDA含量均低于隐睾组[(1.11±0.06)%、(1.14±0.07)nmol/mgprot;(1.73±0.06)%、(2.72±0.13)U/rag]。结论及时补充外源性GSH通过提高睾丸组织的抗氧化能力,进而减少隐睾所致的生精细胞凋亡。Objective To explore the mechanism of glutathione decreasing spermatogenic cell ap- optosis in the rats with cryptorchid. Methods Thirty healthy male SD rats were randomly divided into 3 groups : control group, cryptorehid group and glutathione (GSH) treatment group. The 3 - week tail - sus- pending method was used to establish the rat cryptorchid model. Spermatogenic cell apoptosis index (AI) was detected by terminal - deoxynucleoitidyl transferase mediated d - UTP nick end labeling (TUNEL). The biochemical parameters [ GSH - Px, total antioxidant capacity ( T - AOC ) , and malondialdehyde (MDA) ] in testes were determined by chemical colorimetry. Results The testes weight, GSH-Px activity and T-AOC in the cryptorchid group were lower significantly than in the control group [ (0. 577 ±0. 019) g, (22.68±1.22) U/mg prot, (11.9 ±1.4) U/mg; (1.804±0.022) g, (31.68±0.85) U/mg prot, (29. 7 ±2. 5) U/mg] While the spermatogenic cell A1 and MDA concentration were higher significantly than in the control group [ ( 1.73 ± 0. 06) %, ( 2. 72 ± 0. 13 ) nmol/mg prot ; (0. 55 ± 0. 05 ) %, (0. 53 ± 0.07) nmol/mg prot ], respectively. The testes weight, GSH-Px activity and T-AOC in the GSH treatment group were higher significantly than in the cryptorchid group [ ( 1. 285 ±0. 012) g, (32. 83 ±0. 95) U/mg prot, (30.9±1.8) U/mg; (0.577±0.019) g, (22.68 ±1.22) U/mgprot, (11.9±1.4) U/mg], While the spermatogenic cell AI and MDA concentration were lower significantly than in the eryptorchid group [ ( 1.11 ±0. 06) %, ( 1.14 ±0. 07) nmol/mg prot; ( 1.73 ±0. 06) %, (2. 72 ±0. 13) U/mgJ, re- spectively. Conduslon Timely supplement of exogenous GSH could decrease the spermatogenic cell ap- optosis due to eryptorchid through enhancing the antioxidant ability of testes.
