摘 要:目的观察羟考酮对神经病理性痛大鼠脊髓背角c—fbs蛋白表达的影响。方法成年雄性sD大鼠54只,体重180~250g,随机分为5组(n=18):假手术组(S组)、神经病理性痛组(NP组)、羟考酮+神经病理性痛组(OX组)。s组仅分离坐骨神经但不结扎;NP组采用坐骨神经慢性压迫损伤法制备神经病理性痛模型;OX组于结扎坐骨神经后至处死每天腹腔注射羟考酮15ug/kg,1次/日,连续14d,S组和NP组以等量生理盐水替代。于结扎前1d、结扎后3、7、14d时测定机械痛阈(MWT)和热痛阈(TwL),并于结扎后14d时测定痛阈后处死大鼠,取L4和L5脊髓背角,采用Westernblot法测定c—fos蛋白表达,采用反转录-聚合酶链反应(RT—PCR)法测定c—fosmRNA表达。结果与S组比较,术后3—14d时MWT在NP组[(5.1±1.2)、(4.7±1.6)、(5.2±1.4)g]和OX组[(8.3±1.5)、(9.1±1.8)、(8.7±1.6)g]降低,TwL在NP组[(8.5±1.9)、(6.7±1.6)、(7.2±1.8)S]和OX组[(12.1±2.2)、(11.4±1.9)、(12.5±2.1)S]缩短,术后14d脊髓背角c—fos蛋白的表达在NP组(0.92±0.18)和OX组(0.68±0.15)上调(p〈0.05),c-fosmRNA表达在NP组(1.8±±0.32)和O组(1.33±0,15)升高(P〈0.05);与NP组比较,OX组术后3—14dMWT[(8.3±1.5)、(9.1±1.8)、(8.7±1.6)g]升高,TWL[(12.1±2.2)、(11.4±1.9)、(12.5±2.1)s]延长,术后14d脊髓背角c—fos蛋白的表达(0.68±0.13)下涮,c-fos mRNA表达(1.33±0.15)下调(P〈0.05)。结论羟考酮减轻大鼠神经病理性痛的机制与其下调脊髓背角c-fos表达有关.Objective To investigate the effects of oxycodone on the expression of c - los protein in spinal dorsal horn in a rat model of neuropathic pain (NP). Methods Fifty - four adult male Wistar rats, aged 6-8 weeks, weighing 180-230 g, were randomly divided into 3 groups (n=18 each): sham operation group (S group), NP group and oxycodone group (OX group). In NP group and OX group, 4 ligatures were placed on the right sciatic nerve at 1 mm intervals with 4 - 0 silk thread according to the method described by Bennett et al. In S group, the right sciatic nerves were exposed, but not ligated. In OX group, oxycodone ( 15 p±g/kg) was injected intraperitoneally once a day starting from the end of opera- tion until one day before the animals were sacrificed, while the rats received the equal volume of normal sa- line (5 ml/kg) instead of OX in S group and NP group. One day before operation and 3, 7, 14 days after operation, the mechanical paw withdrawal threshold (MWT) and thermal paw withdrawal latency (TWL) were measured. Animals were sacrificed after measurement of the pain threshold at 14th day after opera- tion, the IA, 15 segment of the spinal cord was removed for determination of c - fos protein expression by Western blotting, and c - fos mRNA expression by reverse transcriptase - polymerase chain reaction (RT -PCR) in the spinal dorsal horn. Results As compared with S group, MWT was significantly de- creased inNPgroup [(5.1 ± 1.2), (4.7 ± 1.6), (5.2 ±1.4) g] and OX group [(8.3 ± 1.5), (9.1 ±1.8), (8.7±1.6) g] at3rd, 7th, and 14thday (P〈0.05), TWLwas shortened in NPgroup [(8.5±1.9), (6.7±1.6), (7.2±1.8) s] andOXgroup [(12.1 ±2.2), (11.4±1.9), (12.5±2. 1 ) s ] at 3rd, 7th, and 14th day ( P 〈 0. 05 ), and the expression of c - fos protein and mRNA was up - regulated in spinal dorsal horn at 14th day after operation ( P 〈 0. 05 ).
