异丙酚对大鼠大脑皮层NR2B蛋白及iNOS mRNA表达的影响

摘  要:为研究异丙酚对大鼠大脑皮层NR2B蛋白及iNOS mRNA表达的影响,选取18只W istar大鼠随机分为3组:生理盐水对照组(C组)、异丙酚麻醉组(P组)和异丙酚麻醉恢复组(R组)。应用免疫组织化学和免疫印迹方法检测大脑皮层NR2B蛋白的表达,进行定位和定量分析。应用实时荧光定量PCR方法检测iNOS mRNA的表达,进行定量分析。结果显示,NR2B蛋白主要在大脑皮层的外颗粒层、外锥体细胞层和内颗粒层的细胞膜上表达。大鼠在输注异丙酚后NR2B蛋白的表达显著下降(P<0.05);iNOS mRNA的表达极显著下降(P<0.01)。结果表明,异丙酚产生麻醉镇痛作用的机制可能是通过降低大脑皮层NR2B蛋白和iNOS mRNA的表达来抑制大脑皮层疼痛冲动的传导。In order to investigate the expression of NR2 B protein and iNOSmRNA in cerebral cortex for continuous infusion of propofol in rats,18 Wistar rats were divided randomly into three groups:control group(C group),propofol group(P group),and recovery group(R group).The expression of NR2 B protein was detected with immunohistochemistry and Western-blot method.The expression of iNOSmRNA was detected by real time PCR.The results showed that NR2 B protein was expressed mainly in the outer granular layer,the external pyramidal layer and the inner granular layer in the cell membrane.The expression of NR2 B protein in cerebral cortex was decreased significantly after receiving propofol(P0.05).The expression of iNOSmRNA in cerebral cortex was decreased significantly after receiving propofol(P0.01).These results indicated that the propofol could inhibit the expression of NR2 B protein and iNOSmRNA,and thus relieve pain impulse to the cerebral cortex.
