摘　　要：【目的】评价重组相思子毒素B链蛋白(r ATB)作为相思子毒素疫苗的免疫原性。【方法】以r ATB为免疫原,采用2×2×2×3析因设计方式分别免疫雌性Balb/C小鼠,主因素包括:重组蛋白的剂量、免疫方式、免疫周期、攻毒剂量。每次采用间接ELISA方法检测小鼠血清中Ig G,并进行天然毒素攻毒实验。体外中和实验依据上述所选的免疫方案进行,分析免疫保护效果。【结果】间接ELISA结果表明r ATB蛋白免疫小鼠后血清中能检测到相应的抗AT特异性抗体,Ig G的效价均可达到1:106以上,PBS对照组则没有抗体产生(P<0.05);小鼠攻毒保护实验和体外中和实验结果说明,腹腔注射r ATB在25μg/只的蛋白免疫剂量下,免疫周期为0-14-28-42 d,可以抵抗最高剂量为5×LD50的天然AT毒素中毒,保护率为100%。【结论】r ATB蛋白免疫小鼠后显示出良好的免疫原性,为相关疫苗研究奠定基础。【Objective】To evaluate the immunogenicity of the recombinant abrin toxin B chain protein（r ATB） as a vaccine.【Methods】By 2×2×2×3 factorial design, female Balb/C mice were immunized by the immunogen of r ATB. The main factors included the dose of r ATB, route of immunization injection, immunization interval and dose of native toxin challenge. The indirect enzyme-linked immunosorbent assay（ELISA） was used to measure the serum antibody titer of Ig G in mice. The native toxin challenge experiment was carried out. The in vitro neutralization assay was done according to the above scheme. And the efficacy of passive protection was analyzed.【Results】The results of indirect ELISA showed that the anti-r ATB antibody could be detected in the sera of mice immunized by r ATB. The titers of Ig G reached 1：106or more. In contrast, the antibody response was not found in PBS-vaccinated mice（P〈0.05）. The results of challenge trials with native AT and in vitro neutralization assay confirmed that the r ATB administered by intraperitoneal injection with25 μ g per mouse could elicit a stronger immune response and protect the mice from a dose of native AT toxin up to 5×LD50in the immunization interval of 0-14-28-42 d. Besides, the protection rate was 100%.【Conclusion】The r ATB can trigger a good immune response and protective efficacy in mice, and it may be a promising vaccine candidate against human exposure to abrin toxin.
