摘  要:目的探讨穿心莲内酯(Andro)对前列腺神经内分泌癌细胞增殖及神经内分泌分化的影响。方法Andro处理前列腺癌细胞(PC3),用MTT法检测48 h时Andro作用的最大效应浓度及对PC3细胞增殖的时间依赖性;进一步提取细胞蛋白,通过Western blotting检测神经内分泌分化标志物(NSE、Cg A)表达情况。结果 Andro作用48 h时7.5μmol/L达到最大效应浓度,且随着时间的延长Andro抑制PC3细胞增殖的作用越显著;Andro能明显抑制PC3细胞的增殖,并具有浓度和时间依赖性。Western blotting检测结果显示,NSE、Cg A是PC3细胞中神经内分泌的标志物,Andro能显著抑制NSE、Cg A的表达。结论Andro可抑制PC3细胞增殖及神经内分泌分化。Objective To investigate the effect of andrographolide on the proliferation and neuroendocrine differentiation of prostate cancer PC3 cells. Methods After PC3 cells were treated with andrographolide,MTT method was used to detect the biggest effective concentration of andrographolide at 48 h and the timedependent relationship at this concentration on PC3 cell proliferation. Meantime,western blot was employed to measure the expression of neuroendocrine differentiation markers NSE and Cg A in PC3 cells. Results Andrographolide significantly inhibited the proliferation of PC3 cells at the dosage of 7.5 μmol / L in a timedependent manner. The expression of NSE and Cg A were decreased in PC3 cells when treated with andrographolide. Conclusion Andrographolide can inhibit the proliferation and neuroendocrine differentiation of PC3 cells.
