摘  要:目的建立反相高效液相色谱法测定舒肺合剂中甘草酸铵含量的方法。方法以Agilent Zorbax SB-C18柱(4.6 mm×150 mm,5μm)为色谱柱,乙腈-0.0025 mol/L庚烷磺酸钠液-0.05 mol/L磷酸二氢钾液(20∶45∶45)(用20%氢氧化钠液调节p H值为7.2±0.05)为流动相,流速1.0 m L/min,检测波长250 nm,进样量10μL,柱温30℃。结果甘草酸铵与相邻组分峰分离良好,阴性样品无干扰。甘草酸铵浓度在23.6~118.1μg/m L之间与峰面积呈良好线性关系,回归方程Y=0.1133X-0.00110,r=0.999 8,加样回收率(n=6)为97.8%,RSD=0.88%。3批舒肺合剂样品测定甘草酸铵含量范围在0.2532~0.2865 mg/m L之间,平均为0.2721mg/m L。结论本法操作简便、准确,可作为舒肺合剂的含量测定方法。Objective To establish a RP-HPLC method for determination of ammonium glycyrrhizinate in Shufei Mixture. Methods The sample was analyzed on an Agilent Zorbax SB-C18column( 4. 6mm × 150 mm,5 μm),and acetonitrile-0. 0025 mol / L sodium heptanesulfonate solution – 0. 05 mol / L potassium dihydrogen phosphate solution( 20 ∶45 ∶45)( adjusted p H value of 7. 2 ± 0. 05 with 20% sodium hydroxide solution) was used as mobile phase. The flow rate was at 1. 0 m L / min. The detective wavelength was at 250 nm. The column temperature was 30 ℃. Results With this chromatographic condition,the ammonium glycyrrhizinate peak in Shufei Mixture sample chromatogram could be separated with other ingredient peaks completely. The negative sample had no interference. The calibration curve was linear at a ranges of 23. 6-118. 1 μg / m L for ammonium glycyrrhizinate,and equation of regression was Y = 0. 1133 X – 0. 00110,r = 0. 999 8. The average recovery from sample was 97. 8% and RSD was 0. 88%( n = 6). The content range of ammonium glycyrrhizinate in three batch Shufei Mixture sample was 0. 2532-0. 2865 mg / m L,and average content was 0. 2721 mg / m L.Conclusion This method is simple,accurate,and useful for control method of this preparation.
