参松养心胶囊对心律失常大鼠抗室性心律失常作用及其机制

摘　　要：目的探讨参松养心胶囊对抗室性心律失常作用及机制。方法氯化钙致大鼠心律失常和缺血再灌注致大鼠心律失常,采用细胞膜片钳技术研究参松养心胶囊对心律失常大鼠L-型钙电流(ICa-L)和瞬时外向钾电流(Ito)的抑制作用。结果 1参松养心胶囊可显著推迟氯化钙致大鼠室性心动过速出现的时间(P<0.01),显著缩短室性心电图的恢复时间(P<0.01),并且作用具有剂量依赖性;2参松养心胶囊中剂量和高剂量可显著降低室性心动过速,室性早搏及心室颤动的发生率(P<0.05或P<0.01);3参松养心胶囊可降低ICa-L电流,使ICa-L电流峰值降低约20%,使ICa-L I-V曲线上移;4参松养心胶囊可降低Ito电流,使Ito峰值降低约30%,使Ito I-V曲线下移。结论参松养心胶囊具有抗室性心律失常的作用,并且对心室肌细胞的ICa-L和Ito具有抑制作用。Objective To study the antiarrhythmia effect of Shensong Yangxin Capsules on arrhythmia rat and its mechanism. Methods The arrhythmia rat were induced by Ca Cl2 and ischemia reperfusion,and the whole-cell patch clamp technique was used to investigate the inhibitive effect of Shensong Yangxin Capsules on ICa-L and Ito. Results 1 Shensong Yangxin Capsules significantly retarded the occurrence of ventricular arrhythmia induced by Ca Cl2（ P 0. 01）,and significantly shortened the recovery time of abnormal ECG（ P 0. 01）with dose-dependence manner. 2 The middle and high dosage of Shensong Yangxin Capsules significantly reduced the incidence rate of ventricular tachycardia,ventricular premature beat,and ventricular fibrillation（ P 0. 05 or P 0. 01）. 3Shensong Yangxin Capsules decreased ICa-L,and made the peak value of ICa-L decreased approximately 20%; Shensong Yangxin Capsules made the I-V curve of ICa-L shift up. 3Shensong Yangxin Capsules decreased Ito with the peak value of ICa-L approximately decreased 20%,and I-V curve of ICa-L shift up; 2Shensong Yangxin Capsules decreased Ito with the peak value of Ito approximately decreased 30%,and I-V curve of Ito shift down. Conclusions Shensong Yangxin Capsules have the antiarrhythmia effect and can inhibit ICa-L and Ito in ventricular myocytes.
