摘　　要：目的:探讨表达HN蛋白的重组腺病毒Ad-HP在体内外对人胃癌细胞的抑制作用。方法:通过MTS法检测Ad-HP(Ad-HN-PEG3p-E1a)对SGC7901细胞的增殖抑制效果,采用Annexin V染色、Caspase酶活性检测、Western blot免疫印迹检测揭示Ad-HP对SGC7901细胞的抑制方式和途径。对BALB/c裸鼠进行瘤内注射Ad-HP探究其在体内的抗肿瘤作用。结果:体外实验中Ad-HP明显抑制SGC7901细胞增殖,并表现出时间效应和剂量效应关系。Ad-HP能提高Caspase酶活性,并释放细胞色素C。体内实验中Ad-HP能显著延缓瘤体生长并延长模型动物的平均生存期,而且能上调IL-2和IFN-γ等细胞因子,使免疫反应朝向Th1优势。结论:Ad-HP可特异性有效抑制胃癌细胞增殖和肿瘤生长。Objective： To investigate the anti-tumor effects of a recombinant adenovirus Ad-HP that express HN protein on human gastric cancer cell proliferation in vitro and in vivo. Methods： In vitro,infection with Ad-HP could inhibit the proliferation of SGC7901,cell viability was assessed by MTS assay,Annexin V assay,Caspase assay and Western blot were used to reveal the suppression apoptosis method and pathway of Ad-HP on SGC7901 cells. In vivo,BALB/c nude mice were treated via intratumoral injection of Ad-HP to investigate the anti-tumor effects. Results： In vitro,Ad-HP significantly suppressed SGC7901 proliferation,and depended on the treat time and treat dose. The infection of Ad-HP resulted in the upregulation of caspase enzymes,and release of Cytochrome C. In vivo,Ad-HP could significantly delay tumor growth and prolong the mean survival of model animals; Ad-HP upregulated the cytokines such as IL-2 and IFN-γ prompt immune reaction to Th1. Conclusion： The oncolytic adenovirus Ad-HP could inhibit proliferation of gastric cancer cells and growth of tumors.
