摘 要:目的:探讨五加双参片对60Coγ射线辐射损伤小鼠的保护作用以及对血清G-CSF水平的影响。方法:将60只小鼠随机分为正常对照组、模型对照组、五加双参片高剂量组、五加双参片中剂量组和五加双参片低剂量组,每组各12只。五加双参片各剂量组给小鼠灌胃给药,剂量分别为6 g·kg^(-1),3 g·kg^(-1)和1.5 g·kg^(-1),正常对照组及模型对照组小鼠灌胃给予0.2 m L生理盐水,每天给药1次,连续灌胃给药10 d。除正常对照组外,各组小鼠于灌胃给药第7 d开始,用60Coγ射线对实验小鼠进行一次性全身照射制备辐射损伤动物模型。结果:五加双参片各剂量组WBC较模型对照组高,尤以五加双参片中剂量组及高剂量组较显著,差异均有统计学意义(P<0.01);五加双参片各剂量组小鼠外周血RBC、PLT、Hb与模型对照组比较无显著差(P>0.05)。与正常对照组比较,模型对照组小鼠遭受辐射损伤后血清中G-CSF表达水平显著降低(P<0.01)。与模型组比较,五加双参片各剂量组小鼠血清中G-CSF表达水平均明显升高,其中高剂量、中剂量组有显著性差异(P<0.01)。结论:五加双参片对60Coγ辐射造血损伤模型小鼠造血粒细胞集落刺激因子和外周血白细胞降低具有显著的拮抗作用。Objective: To discuss the protective effects of Wujia Shuangshen Tablets on the mice with 60Coγradiation injury and its influence on the level of serum G-CSF. Methods: 60 mice were randomly divided into normal control group,model control group,high dosage,medium dosage and low dosage of Wujia Shuangshen Tablets groups,with 12 mice in each group. Intragastric administration of Wujia Shuangshen Tablets with dosages of 6 g·kg-1,3 g·kg-1and 1. 5 g·kg-1was conducted for the mice in the three Wujia Shuangshen Tablets groups respectively; the mice I normal control group and model control group were performed intragastric administration of 0. 2ml normal saline,once daily and 10 days of continuous medication. Except normal control group,other groups were performed one-time whole-body exposure to 60Coγradiation from the 7th day of administration to prepare radiation injury model. Results: The levels of WBC of the three groups of Wujia Shuangshen Tablets were higher than the model control group,especially the medium and high dosage groups were more remarkable; the difference had statistical significance( P 〈0. 05). The levels of RBC,PLT and Hb in the peripheral blood of the three groups of Wujia Shuangshen Tablets had no significant difference with that of model control group( P 〉 0. 05). Compared with normal control group,the expression level of serum G-CSF in model control group after radiation injury markedly declined( P 〈 0. 01). Compared with model group,the expression level of serum G-CSF in the three groups of Wujia Shuangshen Tablets after radiation injury obviously increased,and the medium and high dosage groups had significant difference( P 〈 0. 01). Conclusion: Wujia Shuangshen Tablets has remarkable antagonism against the serum G-CSF and the decline of peripheral WBC in mice with 60Coγradiation injury.
