摘 要:水稻ZH-ct为ZH11在组织培养过程中发现一株突变单株,具有较好的苗期耐冷性,通过人工气候箱低温处理,应用蛋白质双向凝胶电泳(2-DE)检测其低温处理后的差异蛋白表达,并对进行分析。结果显示,共发现差异蛋白468个,对其中重复性好,表达量较高的14个蛋白进行质谱分析,除2个没有检测到蛋白信号外,其余12个分别为光合作用、能量代谢、渗透压调节、过氧化物酶等功能蛋白,为下一步进行耐冷的分子基础及反向遗传学分析提供了基础。ZH-ct possessed the characters of good cold tolerance, which was a mutation plants from tissue culture of zhl 1 callus. Its differential protein expressions in the artificial climate chamber after low temperature treatment by 2-DE were studied. The result showed that there were 468 differential expression proteins spots signed by it' s relative software, in which 14 spots had a better replication and were easily to slice, which were further analyzed by mass spectra, and all of them, except 2 spots were no signs, were photosynthesis, supersession, os- motic pressure adjust, peroxide enzyme, respectively. The results would provide the foundation for rice seeding cold tolerance research and reversed genetics.
