二仙汤对乳腺癌细胞转移大鼠Cdc42的影响

摘　　要：目的:观察二仙汤对乳腺癌细胞转移大鼠细胞分裂周期蛋白42(cell division cycle42,Cdc42)的表达影响,分析其抗乳腺癌转移的作用机制。方法:SD雌性大鼠60只,共分为3组,分别为二仙汤治疗组、5-氟尿嘧啶组、生理盐水组,每组大鼠20只。给药4 d后处死大鼠采取药物血清。乳腺癌HCC1937细胞株培养后分3组,分别加入终体积分数为10%各组血清,处理细胞。统计并检测各组药物血清处理培养后的细胞数目,肿瘤细胞的侵袭能力以迁移细胞的相对数目来表达。另取处理后的各组细胞检测细胞内Cdc42蛋白的水平。结果:二仙汤组对肿瘤细胞的侵袭能力抑制作用明显,与生生理盐水组比较,差异有统计学意义(P<0.05)。二仙汤组细胞内Cdc42水平明显降低,与5-氟尿嘧啶组及生理盐水组比较,差异有统计学意义(P<0.05)。结论:二仙汤有抑制乳腺癌细胞转移的作用,机制可能与抑制体外乳腺癌细胞Cdc42的表达有关。Objective： To observe the influence of Erxian Decoction on the expression of cell division cycle protein 42（ Cdc42） of breast cancer metastasis rat and analyze the mechanisms of anti-cancer metastasis. Methods： 60 SD female rats were randomly divided into three groups,namely Erxian decoction group,5-fluorouracil group and saline group,with 20 rats in each group. The rats were sacrificed 4d after taking drugs and serum was obtained. Cultured HCC1937 breast cancer cell lines were divided into three groups,and three serums of different concentration with final volume fraction of 10% of were added during the cells culture. The number of cells in each group was detected after the serum treatment. The invasive ability of tumor cells was presented by the relative number of migrating cells. Besides,the intracellular Cdc42 protein of cells in each group was detected. Results： The inhibiting effect of Erxian Decoction on the invasiveness of tumor cells in each group was significant compared with that of the raw saline group（ P 〈 0. 05）. The expression of Cdc42 in cells of Erxian soup significantly reduced compared with that of 5-fluorouracil group and the saline group,with the difference being statistically significant（ P 〈 0. 05）. Conclusion： Erxian Decoction can inhibit breast cancer metastasis,and the mechanism may be related to its inhibition effect of Cdc42 expression in breast cancer cells.
