摘  要:为了寻找新型几丁质酶编码基因,提取了汕头湾海域海底表层沉积物中微生物的宏基因组,采用PCR-DGGE技术扩增和分离获得了新型几丁质酶基因编码信息。实验共得到63条几丁质酶基因片段,其编码的蛋白序列与NCBI数据库收录的序列相似率在41%-97%之间,且大多数在70%以下,只有8条相似率在70%以上,NCBI数据库中与其相似性最高的几丁质酶蛋白序列有18个种属,其中与橙色滑柱菌株(Herpetosiphon aurantiacus)ATCC 23779的几丁质酶存在着最高相似性的序列有27条,另外,与未可培养细菌几丁质酶基因片段相似的有22条,占34.9%,分属9个种属。说明汕头湾表层沉积物中存在多种几丁质酶编码基因,其中有很多是尚未研究的,为寻找新型的几丁质酶编码基因提供了有力的生物资源。In order to find new chitinease-encoding genes,we extracted metagenome from surface sediments of the Shantou Bay,and amplified 63 chitinase coding gene fragments using PCR-DGGE. Compared these fragments to NCBI records,the similarity ratios were from 41% to 97%. The most similarity ratios were below 70%,only 8 higher than 70%,and there were 18 species in NCBI chitinase records had the highest similarity ratios. 27 fragments had the highest similarity ratios to the chitinase sequence of Herpetosiphon aurantiacus ATCC 23779. 22 fragments had the highest similarity ratios to uncultured bacteria chitinase sequences which belonged to 9 different species,accounted for 34.9%. The results suggested that there were many different chitinase-coding genes in Shantou Bay sediments,most of them hadn't yet been studied from now. The paper provided a biological resources for finding new chitinease-encoding genes.
