摘  要:目的:观察牛膝多糖对过氧化氢(H2O2)诱导人脐静脉内皮细胞(HUVEC)损伤的保护作用。方法:体外培养HUVEC,分为空白对照组、模型对照组、牛膝多糖低、中、高浓度组(浓度为5%、10%、15%),显微镜观察细胞形态;微板法检测细胞上清液中乳酸脱氢酶(LDH)的活性及一氧化氮(NO)的含量,ELISA法检测内皮素-1(ET-1)、组织型纤溶酶原激活物(t PA)、纤溶酶原激活物抑制剂(PAI-1)的含量。结果:与模型对照组比较,牛膝多糖中、高浓度组细胞形态改善,LDH活性显著减低、ET-1和PAI-1含量显著减少,NO和t PA显著升高。结论:牛膝多糖对H2O2诱导的HUVEC损伤具有显著地保护作用。Objective: To observe the protective effect of Achyranthes bidentata polysaccharideson the injury of human umbilical veinendothelial cell(HUVEC) injured by hydrogen peroxide. Methods: The HUVEC were cultured vitro and divided into five groups as follows: control group, model group, low-dosage of Achyranthes bidentata polysaccharides, mid-dosage of Achyranthes bidentata polysaccharides group and high-dosage of Achyranthes bidentata polysaccharides. Cell morphology was observed under inverted phase contrast microscope. The contents of lactate dehydrogenase(LDH), nitric oxide(NO) were measured by the micro-plate and the method of enzyme-linked immunosorbent assay(ELISA) was used to determine endothelin-1(ET-1), tissue plasminogen activator(t PA) and plasminogen activator inhibitor-1(PAI-1). Results: Compared with the model group, the mid-dosage of Achyranthes bidentata polysaccharides and high-dosage of Achyranthes bidentata polysaccharides significantly improved HUVECs morphology, decreased the contents of LDH, ET-1, PAI-1 and increased the contents of NO and t PA(P〈0.05 or P〈0.01). Conclusion: Achyranthes bidentata polysaccharideshas significant protective effect on HUVEC injured by H2O2.
