摘  要:目的探讨三七总皂苷(PNS)对家兔主动脉粥样硬化的抑制作用。方法将新西兰大白耳兔随机分为对照组、动脉粥样硬化(AS)模型组、PNS中及高剂量治疗组,实验组、治疗组喂食高胆固醇饲料,PNS中、高治疗组分别采取天由兔耳静脉注射PNS50、100 mg·kg·d,每月后2周每天给药3个疗程。第12周末经耳缘静脉采血,检测血清中脂质水平,并取主动脉观察光镜和电镜病理学改变。结果与AS组比较,PNS高、中治疗组兔血清甘油三酯、总胆固醇、低密度脂蛋白胆固醇降低(P<0.05),高密度脂蛋白胆固醇显著增高(P<0.05);光镜下PNS高、中治疗组内膜的泡沫细胞数显著减少。电镜下超微结构观察:AS模型组血管泡沫细胞及中膜平滑肌细胞质内大量脂滴空泡,甚至形成脂囊;PNS中、高剂量组AS病变较AS模型组显著减轻,血管内皮细胞质内见少许小脂滴空泡,各种细胞器存在,内弹力膜存在,中层平滑肌细胞质内偶见脂滴空泡。结论 PNS具有抑制AS形成的作用。Objective: To study the inhibition effect of Panax Notoginseng Saponins( PNS) on artery atherosclerosis in rabbits.Methods: New Zealand white rabbits were randomly divided into a control group,an atherosclerosis( AS) model group and PNS medium- dose and high- dose groups. The experimental group and the treatment group were fed with high cholesterol diet while the PNS medium- dose and high- dose groups were given ear vein injection,PNS50,100 mg / kg / d and administered three times a day at the last two weeks of each month. At the end of the 12 th week,the ear vein blood was taken to detect their serum lipid levels and the aorta was taken to observe their pathological changes by light microscope and electron microscope. Results: Compared with the AS group,the serum triglycerides,total cholesterol and low- density lipoprotein cholesterol of the PNS high- dose and medium- dose groups reduced( P 0. 05)while their high density lipoprotein cholesterol significantly increased( P 〈0. 05). Under the light microscope,the number of foam cells in the intima of the PNS high- dose and medium- dose groups significantly reduced. Under the ultrastructural observation of the electron microscope,there was a large number of lipid droplets and vacuoles,and even the formation of vesicle in the vascular foam cells and the tunica media smooth muscle cytoplasm of the AS model group. The AS lesions of the PNS high- dose and medium- dose groups significantly reduced when compared with that of the AS model group. A few of small lipid droplet vacuoles could be seen within vascular endothelial cytoplasm,with the existence of various cell organelles and elastic membrane. Occasionally the lipid droplets vacuoles could be seen within the tunica media smooth muscle cytoplasm. Conclusion: PNS has an inhibition effect on AS formation.
