摘  要:目的比较相同剂量STZ处理时C57BL/6J和昆明小鼠1型糖尿病模型的成模率及稳定性,探究经济有效的糖尿病小鼠建模品系。方法健康雄性C57BL/6J和昆明小鼠分别随机分为3组(20只/组):正常对照组、150 m g/kg组(单次腹腔注射)、50mg/kg组(连续注射5 d)。末次注射STZ后每天测量小鼠摄食量和饮水量,每周测量小鼠空腹血糖及体重一次,连续观察4周。结果 2种建模方法均能使C57BL/6J和昆明小鼠表现出糖尿病的"三多一少"症状,且空腹血糖值比对照组显著增加(P<0.01),但2种小鼠的血糖差异无统计学意义。实验结束时,C57BL/6J和昆明小鼠的成模率没有差别,50 mg/kg组均为80%,150 mg/kg组均为65%。C57BL/6J小鼠的死亡率略低于昆明小鼠。结论相同剂量STZ处理下C57BL/6J和昆明小鼠1型糖尿病的成模率和模型稳定性基本一致,昆明小鼠是较C57BL/6J小鼠更为经济有效的建立糖尿病模型的小鼠品系。Objective To investigate an efficient and cost-effective mice strain for the establishment of STZ-induced type 1 diabetes model by comparing the molded rate and the stability of mice model of C57 BL /6J and KM mice. Methods Healthy male C57 BL /6J and KM mice were randomly divided into three groups( 20 mice in each group),normal control group,150 mg / kg group( single injection of 150 mg / kg STZ) and 50 mg / kg group( injected for 5 consecutive days),respectively. After the last injection,daily food and water intake were tested,fasting blood glucose level and body weight were examined once a week for 4 consecutive weeks. Results C57 BL /6J and KM mice in both of the groups of 50 mg / kg and 150 mg / kg showed the typical performance of diabetes. The fasting glucose levels in the model groups increased significantly than that of the control groups( P〈0. 01),but no significant differences were observed between the glucose of the two strains of mice. At the end of study,the molded rate of C57 BL /6J and KM mice showed no difference,they were both 80% in the groups of 50 mg / kg and 65% in the groups of 150 m / kg. The mortality rate of C57 BL /6J mice was lower than that of the KM mice,but no significant difference between the two mice strains. Conclusion The molded rate and the stability of mice model of C57 BL /6J and KM mice showed no differences under the same STZ treatment. Thus,the KM mice should be the most efficient and cost-effective strain for the establishment of type 1 diabetic mice model.
