铁皮石斛茎段离体培养一次成苗技术研究

摘　　要：【目的】简化铁皮石斛繁育流程,缩短育苗周期.【方法】以铁皮石斛茎段为外植体,研究了铁皮石斛茎段组织培养一次成苗技术.【结果】70%的乙醇30s+1‰升汞10min消毒效果较好,外殖体污染率为43%,诱导率达97%;一次成苗培养基1/2 MS+NAA 0.5mg/L+6-BA 3mg/L+香蕉泥100g/L+琼脂7‰+蔗糖25g/L增殖及生长情况最好;在该组培系统中铁皮石斛适宜的继代周期为50d,成苗率100%,平均根数5.4,繁殖系数5.6;铁皮石斛丛生芽移栽以腐叶土和腐叶土∶河沙(1∶1)两种基质的成活率高,且生长健壮.【结论】本技术简化了铁皮石斛组培工艺流程,缩短了育苗周期,降低了生产成本,适用于规模化生产.【Objective】This study aim was to simplify the fast-propagation procedure and short the seedling culture time.【Method】The stem section of Dendrobium candidum was used as experiment material,the technology on direct regeneration seedlings was studied by fast-propagation.【Result】The optimum disinfection method was surface sterilization seedlings with 70% ethanol for 30 s,and 1‰ mercuric chloride for 10 min.Optimum seedling cultivation medium was 1/2MS＋NAA 0.5mg/L＋6-BA 3mg/L＋banana puree 100g/L＋agar 7‰＋sugar 25g/L.Meanwhile,the suitable cycle of subculture in fixed was50 days.The rooting rate reached 100%,the mean number of roots reached 5.2,and the effective propagation coefficient reached 5.6after 45 days.Peat and peat river sand（1∶1）were the good transplantion matrix.【Conclusion】It is suggested that the method shorts the cycle of seedling cultivation,lows the production cost.
