摘  要:目的探讨丙泊酚对新生小鼠大脑梨状皮层神经前体细胞增殖的影响。方法选取健康同窝、日龄为7 d(P7)的C57小鼠15只,按照随机数字表法分为3组:对照组,丙泊酚低剂量(30 mg/kg)组,丙泊酚高剂量(60 mg/kg)组,所有小鼠从P7开始接受药物处理。高、低剂量组分别腹腔注射丙泊酚60、30 mg/kg,对照组腹腔注射相同体积10%脂肪乳。药物处理后24 h(P8)腹腔注射Brd U 50 mg/kg,2 h后取材,采用免疫组织化学法检测梨状皮层细胞增殖标记物Brd U、Sox2及神经干细胞纤维突起标记物Nestin的表达。结果与对照组(42.67±2.41)比较,丙泊酚低剂量组(33.84±1.33)及高剂量组(25.17±1.73)新生小鼠大脑梨状皮层中Brd U+细胞数量明显减少;与低剂量组比较,高剂量组Brd U+细胞数量减少更明显(P<0.05);免疫组化结果显示:丙泊酚低剂量组Sox2标记的神经祖细胞数(558.94±29.85)较对照组(618.67±24.01)差异无统计学意义(P>0.05),但丙泊酚高剂量组(430.83±29.23)较对照组、丙泊酚低剂量组均减少(P<0.01,P<0.05)。丙泊酚低剂量组(24.73±0.69)与对照组(34.81±2.56)比较,Brd U-Sox2双标阳性细胞数量差异无统计学意义(P>0.05);但丙泊酚高剂量组(12.48±0.80)较对照组及丙泊酚低剂量组均减少(P<0.01,P<0.05)。免疫荧光染色结果显示:与对照组(1.00±0.00)比较,丙泊酚低剂量组(0.65±0.07)及高剂量组(0.60±0.08)Nestin标记的神经干细胞纤维突起灰度值降低,纤维分布稀疏且排列紊乱(P<0.01)。结论丙泊酚可抑制新生小鼠大脑梨状皮层神经前体细胞增殖,并呈一定剂量相关性。Objective To determine the effect of propofol on the proliferation of neural precursor cells( NPCs) of piriform cortex in neonatal mice. Methods Fifteen healthy 7-day-old C57 mice from the same litters were randomly divided into 3 groups,that is,control group,low-( 30 mg / kg) and high-dose propofol groups( 60 mg / kg). The animals of the 2 latter groups were treated with corresponding drugs on postnatal days 7( P7) by intra-peritoneal injection,while those of the control group were injected with the same volume of 10% intralipid. They were given intra-peritoneal injection of 50 mg / kg Brd U on P8,and then were sacrificed in 2 h for brain tissue. Immunohistochemical assay was used to detect the expression of Brd U,Sox2 and Nestin( neural stem cell marker) in piriform cortex. Results Compared with the control group( 42. 67± 2. 41),the number of Brd U-positive cells in piriform cortex was significantly less in the low-( 33. 84 ± 1.33,P 〈 0. 05) and high-dose propofol groups( 25. 17 ± 1. 73,P 〈 0. 01),with that of the high-dose group obviously decreased than that of low-dose group( P 〈 0. 05). There as no significant change in the number of Sox2-positve cells in piriform cortex of the low-dose propofol group( 558. 94 ± 29. 85) and the control group( 618. 67 ± 24. 01,P 〉 0. 05),but that of the high-dose propofol group( 430. 83 ± 29. 23) was remarkably lower than those of the low-dose( P 〈 0. 01) and control group( P 〈 0. 01). Further more,the number of Brd U-Sox2 double positive cells were not significant decreased after low-dose propofol treatment( 24. 73 ± 0.69) compared with the control group( 34. 81 ± 2. 56,P 〉 0. 05),while the high-dose treatment significantly decreased the number as compared with that in the control group( 12. 48 ± 0. 80,P 〈 0. 01) and the low-dose group( P 〈 0. 05).
