摘  要:目的:检测不同环境对小鼠孕期应激子代海马磷酸化环磷酸腺苷反应元件结合蛋白(pCREB)的影响。方法:将12只孕小鼠随机分为对照组和实验组。对照组动物不做任何处理,其子代断乳后喂养于标准环境;实验组动物在孕第12~18天实施限制性应激,其子代断乳后一半喂养于标准环境(应激+标准环境组),另一半子代喂养于丰富的环境(应激+丰富环境组)。子代出生后5周被处死,用免疫印迹和免疫组织化学显色法检测海马内p-CREB的表达。结果:p-CREB免疫阳性产物广泛分布于海马各区神经元。免疫印迹显示应激+标准环境组p-CREB表达明显少于对照组(P〈0.05);应激+标准环境组海马p-CREB的表达也明显少于应激+丰富环境组(P〈0.05),应激+丰富环境组与对照组比较差异无统计学意义(P〉0.05)。结论:丰富环境能有效改善孕期应激造成的子代海马p-CREB表达降低。Objective:To observe the effects of different environments on the hippocampal phosphorylated cyclic adenosine monophosphate response element binding protein(p-CREB)of antenatal stressed offsprings.Methods:Pregnant mouse were randomly divided into control group and experimental group.The control group received no stress.Their offspring were raised at the standard environment.The experimental group received the restraint stress at pregnancy day 12~18.The half of the offspring were raised at the standard environment(stressed+standard environment group),and the others were raised at the enriched environment after weaning(stress+enriched environment group).The offspring were executed after 5week,the p-CREB expression in the hippocampus was observed by immunohistochemistry and western blotting.Results:The p-CREB was expressed in the hippocampus neurons of mouse.Compared to the control group,the p-CREB expression in hippocampus of stressed group was significantly reduced(P〈0.05).The p-CREB expression of the stressed group was significantly lower than that of the stress+enriched environment group(P〈0.05).Conclusion:Environmental enrichment can efficiently reverse the decline of p-CREB expression in the hippocampus of offsprings caused by antenatal stress.
