摘 要:目的观察人参皂苷Rg3对兔耳病理性瘢痕组织中TNF-α和IL-1β表达的影响。方法建立兔耳病理性瘢痕模型,随机分为治疗组和对照组;各组均用微量注射器行于创面行局部封闭注射,治疗组注射人参皂苷Rg3,对照组注射等量的PBS;通过比较各组瘢痕指数(SEI)及I、III型胶原表达不同评价瘢痕增生程度的差异;采用免疫组化方法检测炎症因子TNF-α和IL-1β的表达。结果与PBS对照组相比较,人参皂苷Rg3治疗组兔耳病理性瘢痕组织SEI及I、III型胶原沉积显著下降,炎症因子TNF-α和IL-1β表达减少,差异具有统计学意义(P<0.05)。结论人参皂苷Rg3在创面愈合早期及时干预,可抑制兔耳增生性瘢痕组织中炎症因子TNF-α和IL-1β的表达,减少瘢痕的形成,提示其抗瘢痕作用可能与抑制炎症反应有关。Objective To observe the effect of Ginsenoside-Rg3 on the expression of interleukin-1β(IL-1β) and tumornecrosis factor-α(TNF-α) in the ear wounds. Methods Collagen I and III expression detected by immunohistochemistry and scar elevation index(SEI)was used to evaluate the extent of scarring. The expression of inflammatory factors interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α) was detected by immunohistochemistry. Results Compared with control scars,GinsenosideRg3-treated wounds exhibited decreased inflammation significantly as evidenced by the lower levels of IL-1β and TNF-α. The SEI and the synthesis of collagens I and III were significantly decreased in the Ginsenoside-Rg3-treated scars(P 〈0.05). Conclusion Immediate topical injection of Ginsenoside-Rg3 during the wound healing process can reduce the severity of hypertrophic scarring in a rabbit model,which may result from controlling inflammation,inducing fibroblast apoptosis and promoting collagen degradation by decreased the inflammatory factors of IL-1β and TNF-α.
