摘  要:目的采用高效液相色谱法对不同连作条件下穿山龙中的皂苷类活性成分进行含量测定,探讨不同连作条件对于穿山龙皂苷类活性成分含量的影响。方法采用RP-HPLC,色谱柱:Agilent TC-C_(18)(4.6 mm×250 mm,5μm);乙腈(A)-水(B)为流动相梯度洗脱;流速1.0 mL/min,柱温:35℃,检测波长:203 nm,计算皂苷类成分含量,并采用SPSS等软件对所得结果进行分析。结果吉林通化种植基地的穿山龙中,除了甲基原薯蓣皂苷为连作一茬3年时含量最高,其他三种皂苷类成分原薯蓣皂苷、纤细薯蓣皂苷、薯蓣皂苷均为正茬4年时最高,而后随着连作年限的增加,含量逐渐减少,并且原薯蓣皂苷与薯蓣皂苷含量呈正相关。结论穿山龙随着连作年限的增加,其皂苷类活性成分的含量也随之下降,说明连作障碍对其皂苷类活性成分的含量积累造成了影响,但正茬4年时其皂苷类成分总含量最高,提示正茬4年为采收最佳时间,这为该药材的质量控制及GAP规范种植提供了依据和示范。Objective HPLC method is used to determine the content of active ingredient saponins in Dioscoreae nipponicae Rhizoma under different conditions of continuous cropping obstacle and to study the effect of continuous cropping obstacle on the content of active ingredient saponins. Methods Samples were achieved and separatedby Agilent TC-C18 column( 250 mm × 4. 6 mm,5 μm) with acetonitrile-water as mobile phase for gradient elution. The flowrate was 1. 0 m L / min. The column temperature was 35 ℃,the UV detector was set at 203 nm,ingredients saponins content was calculated,and SPSS software was used to analyze the results. Results In the Dioscoreae nipponicae Rhizoma,in the plantation base in Tonghua of Jilin province,except M ethyl protodioscin which had the highest content at 3 years,the other three saponins,including protodioscin,diosgenin slim and dioscin had the highest content at four years,and then with the increase of years,the content gradually decreased,and the content of protodioscin and diosgenin was positively correlated. Conclusion With the increase of continuous cropping,the content of active ingredient saponins in Dioscoreae nipponicae Rhizoma decreases,which indicates that continuous cropping obstacle affects the content accumulation of active ingredient saponins. Due to the fact that the content is the highest at four years,4 years is the best time to harvest,which provide the basis and model for the quality control of the medicine and for GAP standerdized planting.
