摘  要:本研究首次在小麦幼胚脱分化过程中利用Affymetrix小麦基因芯片技术对MADS-box基因表达变化进行检测,共检测到20个MADS-box基因,其中14个基因下调表达,6个基因混合表达。这些基因大量下调表达,推测其在小麦幼胚脱分化中具有较强的抑制作用。此外发现,12 h是小麦幼胚脱分化过程中MADS-box基因表达的一个关键时期。选取CA670406、CK213813、CA608865和AB107992四个基因进行半定量RT-PCR验证,检测结果与小麦基因芯片结果基本一致。The expression profiles of MADS-box genes during dedifferentiation of wheat immature embryo were firstly analyzed using wheat gene chip technique. Total 20 MADS-box genes were detected,among which,14 genes were down-regulated and 6 genes presented mixed expression. Down-regulated expression of most of these MADS-box genes indicated that they might have strong inhibitory effects during the differentiation of wheat immature embryo. Besides,the 12 th hour was a key expression period of these MADS-box genes. The expression profiles of 4 genes,including CA670406,CK213813,CA608865 and AB107992,were verified by semi-quantitative RT-PCR technique,which were basically consistent with those of wheat gene chip.
