糖尿病肾病患者染色体端粒长度改变的研究

摘　　要：目的：研究糖尿病肾病患者染色体端粒长度的变化。方法分别取40～50岁组和60～70岁组正常人和糖尿病肾病患者的外周血，分离出白细胞，提取DNA后，采用Southern 杂交对外周血白细胞染色体的端粒DNA序列长度进行检测。结果60～70岁组的端粒长度短于40～50岁组（P＜0．05），两年龄组糖尿病肾病患者的端粒长度短于同年龄组正常人（P＜0．05）。结论糖尿病肾病可使患者外周血白细胞染色体端粒长度缩短，端粒长度的缩短可能是糖尿病肾病的发病机制之一。Objective To investigate the changes of telomere length in diabetic nephropathy patients .Methods Healthy subjects and diabetic nephropathy patients were included into the current study , and then divided into Group forty to fifty year-old and Group sixty to seventy year -old.Their peripheral blood was collected to separate leukocytes for DNA extraction .Then the length of telomere in these leukocytes was measured by Southern blotting .Results Healthy subjects aging sixty to seventy showed shorter telomere than those from forty to fifty year -old （P〈0.05）.Patients suf-fering with diabetic nephropathy had shorter telomere than healthy subjects at matched ages （P〈0.05）.Conclusion These results indicate that diabetic nephropathy may partly contribute to shortened length of telomere in peripheral leuko -cytes.Shortened length of telomere may be one of the mechanisms of diabetic nephropathy .
