摘 要:目的探讨实验室温湿度对高通量酶联免疫吸附实验(ELISA)测定丙型肝炎病毒抗体(HCV-Ab)标准品结果的影响。方法利用艾德康全自动酶免仪(ELISA 1100型),恒温24℃、不同相对湿度(%RH)(30%RH、40%RH,50%RH、60%RH、70%RH、80%RH、90%RH)条件下及湿度50%RH、不同温度(16℃、18℃、20℃、22℃、24℃、26℃、28℃、30℃、32℃、34℃、36℃、38℃、40℃)条件下,分别对HCV-Ab标准品进行测定。结果在排除湿度影响的情况,在温度16℃~36℃条件下进行实验显示,HCV-Ab的S/CO值(分别为3.93±0.09,4.43±0.15,5.16±0.11,6.00±0.17,6.43±0.12,7.43±0.10,7.84±0.15,7.97±0.47,8.24±0.29,8.64±0.61,9.24±0.43)之间差异有统计学意义(F=620.08,P=0.000),且标准品S/CO值均随着温度升高而逐渐升高,HCV-Ab标准品的ELISA结果与温度呈正相关(r=0.97)。36℃和38℃之间S/CO值(9.24±0.43,9.29±0.30)差异无统计学意义(t=0.52,P>0.05),而38℃和40℃之间的S/CO值(9.29±0.30,8.65±0.77)差异有统计学意义(t=8.17,P<0.05)。而在排除温度影响的情况,不同湿度(30%RH、40%RH,50%RH、60%RH、70%RH、80%RH、90%RH)间S/CO值结果(分别为7.20±0.18,7.09±0.32,5.83±0.27,5.38±0.24,3.91±0.28,3.27±0.49,3.04±0.85)差异有统计学意义(F=985.64,P=0.000),且标准品S/CO值随着湿度升高而逐渐降低,HCV-Ab标准品ELISA结果与湿度呈负相关(r=-0.98)。结论不同的温湿度对高通量ELISA检测结果有着重要影响,尽量选择温度不超过36℃和湿度较低的环境进行高通量的ELISA测定,综合考虑推荐全自动酶免仪工作温度范围控制在18℃~25℃之间,湿度控制在35%RH^50%RH之间。Objective To explore the influence of room temperature and humidity on the measurement results of hepatitis C virus antibody using high-throughput ELISA method. Methods To detect the S / CO values of HCV antibody standard using addcare automatic enzyme immunoassay instrument( ELISA 1100)under 24℃ and different humidity( 30% RH,40% RH,50% RH,60% RH,70% RH,80% RH,90% RH) and50%RH and different temperatures( 16℃,18℃,20℃,22℃,24℃,26℃,28℃,30℃,32℃,34℃,36℃,38℃,40℃) respectively. Results The S / CO values of HCV-Ab standard showed significant difference under different temperature( 16℃,18℃,20℃,22℃,24℃,26℃,28℃,30℃,32℃,34℃,36℃),and the S / CO value of standard was positively correlated with temperature. The S / CO value altered little between36℃ and 38℃,but there was of significant difference between 38℃ and 40℃. While the S / CO value of the standard gradually decreased with the rising of humidity( 30%RH,40%RH,50%RH,60%RH,70%RH,80%RH,90% RH),which presented significant difference between different humidity( P〈0.05),and S / CO value of the standard had a negative correlation with the humidity. Conclusions Room temperature and humidity have important effects on high-throughput ELISA detection results. High-throughput ELISA should been carried out in the environment with no more than 36℃ and low humidity.Comprehensive consideration is recommended for the automatic enzyme immunoassay instrument with operating temperature range from 18 to25 degrees,humidity control between 35%RH ~ 50%RH.
