健脑益智汤对阿尔茨海默病大鼠P53的影响

摘　　要：目的研究健脑益智汤对阿尔茨海默病（AD）大鼠海马P53蛋白表达的影响,探讨其对AD细胞凋亡的防治作用机制。方法正常老年大鼠60只,分为对照组、模型组、安理申组、健脑益智汤小、中和大剂量组,采用侧脑室注射聚集态Aβ_（25-35）建立AD模型,药物灌胃4周,Western-blot法检测海马P53蛋白及磷酸化P53（Ser15）蛋白（pP53-Ser15）表达水平。结果与对照组比,模型组P53及p P53-Ser15的表达增多（P〈0.01）;与模型组比,安理申组P53蛋白降低（P〈0.01）,pP53-Ser15降低无统计学意义,健脑益智汤随剂量增加,P53蛋白及pP53-Ser15呈下降趋势（P〈0.05,P〈0.01）;健脑益智汤大剂量组pP53-Ser15下降较安理申组明显（P〈0.05）。结论健脑益智汤降低AD大鼠细胞凋亡的可能机制是通过降低P53及pP53-Ser15蛋白的表达来实现的。Objective To explore the effect of Jiannao Yizhi decoction on hippocampal expression level of P53 and pP53-Ser15 in rats with Alzheimer＇s disease,and to investigate the mechanism of the prevention and treatment of AD cell apoptosis. Methods Selecting normal old rats that were randomly divided into six groups. We used injected accumulation Aβ25-35 in lateral ventricles to make AD models. Each group was given the regulation lavage four weeks. The levels of apoptosis related proteins expression level of P53 were detected by Westernblot. Results Comparing with the control group,the expression of hippocampal apoptosis related proteins P53 and pP53-Ser15 of the model group were increased. Comparing with the model group,P53 protein of the aricept group reduced（ P〈0. 01）,phosphorylation P53（ Ser15） reduced,and there was no statistical significance. And with the increase of Jiannao Yizhi decoction,there were a down-regulation of the protein expression patterns of P53 and pP53-Ser15（ P〈0. 05,P〈0. 01）. Phosphorylation P53 of the Jiannao Yizhi decoction high-dose group significantly decreased than the aricept group（ P〈0. 05）. Conclusions Jiannao Yizhi decoction could reduce the expression level of apoptosis related proteins P53 and pP53-Ser15 in order to increase hippocampal apoptosis of Alzheimer＇s disease.
