摘 要:目的:探讨五味子多糖对H22荷瘤小鼠肿瘤组织病理结构及总蛋白质组的影响。方法:采用光学显微镜与电镜观察的方法,观察五味子多糖对H22实体瘤组织的病理学影响;采用双向电泳及银染法,观察五味子多糖对H22实体瘤组织蛋白质表达差异的影响。结果:五味子多糖给药组细胞异型性降低,癌组织中浸润大量的免疫细胞(白细胞和淋巴细胞),同时,肿瘤组织细胞及胞浆内细胞器都伴有坏死的现象;与正常组比较,五味子多糖高剂量组可使在模型组表达上调的9个蛋白质斑点表达量下降,接近正常组。结论:五味子多糖能抑制小鼠H22肝癌移植瘤的生长,其作用机制可能与下调肿瘤组织中蛋白质表达有关。Objective: To discuss the influence of Fructus schisandrae polysaccharides on tumor tissue pathological structure and total protein group in H22 mice tumor-burdened. Methods: The pathological changes of H22 tumor tissue was observed with light microscope and electron microscope, and the differential expression protein of H22 Tumor tissue with two dimensional electrophoresis and silver staining method. Results:Heterogeneity of tumor tissue cells was reduced; a large number of immune cells(White blood cells and lymphocytes) infiltrated carcinoma and the tumor cells and intracellular organelles were necrosis in the drug group of Fructus schisandrae polysaccharides. The drug group of Fructus schisandrae polysaccharides decreased expression of quantity of 9 protein spots compared with high expression amount in the model group(compared with the normal group) and closed to expression of quantity in the normal group. Conclusion: Fructus schisandrae polysaccharides inhibited the growth of H22 tumor in mice; the mechanism maybe associated with decreasing high expression protein in tumor tissue.
