摘 要:目的:探索咳嗽变异性哮喘(CVA)痰瘀互结病证结合大鼠动物模型的构建方法。方法:模拟中医病因,以幼龄大鼠肺脾气虚,烟熏致痰浊内生,长期激怒联合高脂饮食致气滞血瘀,建立痰瘀互结中医证候模型;模拟西医病因病理,卵蛋白氢氧化铝致敏,辣椒素激发,建立西医大鼠CVA模型。观察大鼠一般状态、自发活动情况、咳嗽反应、肺支气管病理组织形态、支气管肺泡灌洗液(BALF)嗜酸性粒细胞计数。结果:痰瘀互结模型组大鼠体质量增长缓慢(P<0.01),自发活动减少(P<0.01),咳嗽次数增加(P<0.01);支气管管腔狭窄,黏膜增厚,肺泡腔增大,黏液分泌增多,大量嗜酸性粒细胞浸润(P<0.01);与模型组比较,痰瘀同治组各项观测指标均有明显改善(P<0.01)。结论:模拟传统中医病因、西医病因病理建立CVA动物模型;以整体观念、微观验证综合评价中医CVA动物模型;以方测证,痰瘀同治反证CVA动物模型。多因素复合作用可成功复制CVA大鼠病证结合动物模型。Objective: To establish the animal model of phlegm accumulating and blood stasis in cough variant asthma Methods: According to simulating the Traditional Chinese Medicine(TCM) etiology, the young rats with Qi deficiency in lung and spleen in natural were smoked to internal phlegm turbidity. Then the rats were fed with high fat diet and irritated for a long time to create the phlegm and blood stasis syndrome; thus the animal model of phlegm accumulating and blood stasis was formed. According to simulating the western medicine etiology pathology, the young rats were sensitized by egg protein aluminum hydroxide and inspired by capsaicin, and then the animal model of cough variant asthma rat was established. The general state, spontaneous activity, cough response, bronchial lung pathology organization form and bronchoalveolar lavage fluid(BALF) eosinophil count were observed. Results: The rat weight increased slowly in phlegm and blood stasis group(P 0.01); spontaneous activity decreased(P 0.01), and cough frequency increased(P 0.01); The bronchial lumen stenosis, mucosal thickening, alveolar space increased; mucus secretion increased, with a large number of eosinophil infiltration(P〈0.01); Compared to the model group, the observation indexes of treatment on phlegm and blood stasis group were obviously improved(P〈0.01). Conclusion: Multifactor composite effects can be used successfully in replication of animal model of cough variant asthma rat with combination of disease and syndrome, according to simulating the TCM etiology and western medicine etiology pathology, evaluating the animal model on the holism and microcosmic, and proving the model by taking a square to test certificate and treatment on phlegm and blood in opposite.
