摘  要:目的通过对新型碳化硅-碳(C/C-Si C)复合材料进行生物相容性的相关研究,论证其作为人工小关节假体替代材料上的可行性。方法将灭菌处理后的C/C-Si C复合材料制成表面积与浸提介质体积比率分别为1 cm^2/ml、3 cm^2/ml、5 cm^2/ml的C/C-Si C复合材料浸提液作为实验组,观察小鼠成纤维细胞(3T3)在其中的的生长状态,并测定各自吸光度(A值),评定其细胞毒性情况。按50 ml/kg的比重每隔24 h分别腹腔注射3 cm^2/ml的浸提液、环磷酰胺溶液及生理盐水,毒染4次后取股骨骨髓液,制得涂片,观察含有微核的嗜多染红细胞,分析其遗传毒性情况。沿兔耳缘静脉取8 ml新鲜兔血,抗凝处理后水浴1 h,观察并计算溶血率,判定其是否发生溶血反应。将白化豚鼠背部除毛,在其除毛区上半部分别敷盖C/C-Si C材料表面积与浸提介质体积比为3 cm^2/ml、5%甲醛(阳性对照组)和生理盐水(阴性对照组)的纱布6 h,重复3周后去除,14 d后再于除毛区下半部敷盖三种材料,观察24、48 h的皮肤情况并分级。结果生物毒性方面,实验组中细胞生长情况良好,形态正常,且随时间延长细胞数量逐渐增加。遗传毒性方面,骨髓液中嗜多染红细胞的微核数逐渐增加,与生理盐水组无统计学差异(P>0.05)。溶血反应中,实验组与阴性对照组无统计学差异(P>0.05),与阳性对照组有统计学差异(P<0.05)。致敏试验中,实验组仅1例在24 h内呈现1级反应,且在48 h内消失。结论 C/C-Si C复合材料无细胞毒性及细胞遗传毒性,无致敏反应及溶血反应,具有较好的生物相容性,可替代传统的C/C材料成为人工小关节假体的理想材质。Objective To demonstrate the feasibility of C/C-Si C composite materials as a substitute for artificial joint prosthesis based on the research of mechanical properties and biological compatibility of it. Methods The C/C-Si C composite's extract liquid was prepared and the ratio of surface to volume were 1 cm2/ ml,3 cm2/ ml,and 5 cm2/ ml. The 3T3 cells 'growing status was observed by absorbance( A) value and the cytotoxicity was evaluated. The extracted liquid which the ratio of surface to volume was 3 cm2/ ml,cyclophosphamide solution,and saline were intraperitoneally injectied according to the proportion of 50 ml/kg every 24 hours. After four times of the injection,the thighbone marrow fluids smear was obtained,the polychromatic erythrocytes which had micronucleus were observed and the toxicity statue was assessed. Fresh blood of 8 ml was obtained from the rabbit auricular veins,1-hour water bath was conducted after anti-coagulation treatment; the hemolysis was observed to judge whether the hemolytic reaction had happeded. The albino guinea pigs were epilated,and the top half of the depilating area was covered with gauzes soaked by C/C-Si C material extraction( surface to volume was 3 cm2/ ml),5%formaldehyde( positive control group) or saline( negative control group) for six hours; this processe was repeated for three weeks. Three types of material were put on the lower half of the depilating area,the 24-and 48-hour skin statuses were observed and rated. Results In biotoxicity test,the cells in the experimental group grew well with normal morphology,and the number increased gradually with the time extended. In genetic toxicity test,micronucleus of the polychromatic erythrocytes in the marrow fluids also increased gradually and had no significant difference between the experimental group and the saline group( P 〉0. 05).
