HPLC法对白香丹胶囊中芍药苷在大鼠体内药时曲线的测定

摘　　要：目的:本研究通过HPLC法对白香丹胶囊大鼠含药血清的检测,确定其主要药效成分芍药苷在体内代谢的药时曲线,并找出最佳采血时间,为后续机制研究奠定基础。方法:通过方法学考察确定所采用HPLC的准确度和精密度;给予白香丹胶囊溶液1 mL/100 g,连续给药3天,每天2次;给药后在不同时间点对大鼠进行眼眶静脉丛取血,白香丹胶囊组取血时间点分别为:10、20、40、60、90、120、180、240、300、360 min,离心15 min,3 000 r·min^(-1),取血清;将大鼠血清与甲醇按1:4混合,4℃离心,取上清液,氮气吹干,流动相定容至100μL,过滤后进行测定。结果:方法学考察结果显示,日内精密度和日间精密度RSD<3%,回收率均在98.0%-102.0%;制备药时曲线显示末次给药2 h左右含药血清中芍药苷含量达到峰值。结论:通过方法学考察说明所采用HPLC方法日内精密度和日间精密度良好,回收率在标准范围内,符合定量检测要求;采用该方法所得药时曲线说明该复方中主要药效成分芍药苷在2 h后血药浓度最大,可作为复方含药血清提取的最佳采血时间,这将为后续该复方治疗机制研究提供科学依据。The goal of the investigation was to detect the time-concentration curve of paeoniflorin of Baixiangdan capsule in rats in vivo with HPLC,and screen out the best drawing-blood time in order to lay the foundation for further mechanism researches of Baixiangdan capsule.In this study,accuracy and precision of HPLC were determined in accordance with selected methodology inspection.Baixiangdan capsules were dissolved as 1mL/100 g in proportion.Then administration of Baixiangdan solution was given twice a day for continuous three days.At 10,20,40,60,90,120,180,240,300,360 min after administration,1 mL of peripheral blood was collected from fundus oculi venous plexus of rats,and mixed with methyl alcohol by 1：4,centrifuged at 3 000 rpm for 15 min at 4℃.Then serum was collected and dried with N_2 flow,and dissolved at mobile phase.It was found that the RSD for intra- and interday precision of HPLC was less than 3%,while the recovery rate was 98.0%-102.0%.The time-concentration curve showed that the content of serum paeoniflorin reached to the peak at 2h after the last administration.All the results indicated a good condition of intra- and interday precision of HPLC.The recovery rate was in the standard range,and met the requirements of quantitative detection,while the timeconcentration curve suggested that blood concentration of paeoniflorin was at peak at 2 h after administrating Baixiangdan solution,which may provide scientific evidences for compound treatment mechanism studies in the future.
