摘 要:以拟南芥植株为材料,观察了反式-2-己烯醛熏蒸处理后拟南芥初生根生长变化。采用非损伤微测技术,从动态生长素流的角度研究了反式-2-己烯醛处理对拟南芥根尖生长素转运的影响,并结合实时定量PCR技术检测了反式-2-己烯醛处理后生长素极性转运相关基因的表达。结果表明,反式-2-己烯醛处理能够抑制拟南芥初生根生长,且浓度越高抑制作用越强烈。反式-2-己烯醛显著抑制拟南芥根尖分生区及过渡区的生长素外排,并显著下调了生长素极性转运基因PIN1、PIN2、PIN3及生长素感知基因TIR1的表达。说明反式-2-己烯醛抑制拟南芥根尖生长素的极性转运从而影响初生根生长。In this study, primary root length of Arabidopsis thaliana seedlings was measured after E-2-hexenal fumigation. Using non-invasive micro-test technique, IAA flux rate was detected in different functional regions of the root apex with or without E-2-hexenal treatment. Furthermore, the expression levels of several polar auxin transport relevant genes were quantified by quantitative Real-time PCR. The result indicated that the primary root growth of Arabidopsis seedlings was strongly inhibited after E-2-hexenal exposure, and the inhibition was significant along with the increased concentrations of this aldehyde. E-2-hexenal significantly repressed the auxin efflux rate in the meristem and transition zone of seedling roots. E-2-hexenal also down-regulated the transcription expression level of auxin transport-related genes PIN1, PIN2, PIN3, and the IAA perception gene TIR1. Therefore, E-2-hexenal has the negative effect on polar auxin transport in roots, which finally inhibited the primary root elongation.
