摘 要:目的探讨姜黄素预处理对大鼠急性脊髓损伤后环氧合酶-2(COX-2)和诱导型一氧化氮合酶(i NOS)表达的影响。方法成年雌性SD大鼠150只,体重250-300 g,随机分为3组(n=50),分别为假手术组、模型组和姜黄素治疗组。用改良的Allen's法建立急性脊髓损伤模型后,假手术组仅咬除椎板,暴露脊髓。术后1、3、7、14、21 d行BBB运动功能评分及斜板实验评价大鼠后肢功能,然后处死大鼠并取材,进行免疫组织化学染色及Western blot检测,观察COX-2和i NOS的表达变化。结果各时间点模型组和姜黄素治疗组BBB评分及斜板实验评分均显著低于假手术组(<0.05);但是自7d开始,各时间点姜黄素处理组大鼠后肢功能恢复情况明显优于模型组(<0.05)。各时间点假手术组大鼠脊髓组织COX-2和i NOS阳性表达均显著低于模型组和姜黄素治疗组,但姜黄素治疗组显著低于相同时间点模型组(<0.05)。结论姜黄素对脊髓损伤大鼠的神经保护作用可能与下调COX-2和i NOS表达相关。Objective To explore the effect of curcumin on the expressions of cyclooxygenase-2(COX-2)and inducible nitric synthase(i NOS)in rats with acute spinal cord injury.Methods A total of 150 rats,weighing 250-300 g,were randomly divided into 3 groups(n=50):sham-operation group,model group and curcumin treatment group(200-1mg·kg).The animal model was established following improved Allen's model.The Basso,Beattie and Bresnahan(BBB)locomotor rating scale and inclined plane test were used to evaluate rat behavioral consequences at 1,3,7,14,and 21 days after injury.Immunohistochemical staining and Western blot assay were used to observe the expressions of COX-2 and i NOS.Results BBB score and inclined plane test score were significantly lower in model group or curcumin treatment group than in sham group at each time point(p〈0.05),but increased significantly in curcumin treatment group than in model group from 7d to 21 d after SCI.The expression levels of COX-2 and i NOS in spinal cord were lower significantly in sham-operation group than in model group or curcumin treatment group(p〈0.05),however,lower in curcumin treatment group than in model group.Conclusion The protective effect of curcumin on acute spinal cord injury might be related to the decrease of COX-2 and i NOS expressions.
