摘  要:目的研究神经节苷脂(GM1)对急性脊髓损伤大鼠Fas和Caspase1表达的变化。方法选取72只成年健康SD大鼠,按Nystrom法建立大鼠脊髓损伤模型,按随机数字表法分为假手术组24只、脊髓损伤组24只、GM1组24只,脊髓损伤组和GM1治疗组依据脊髓损伤后的不同时间点再细分为1d、7d和14d三个亚组。伤后第1天、第7天和第14天分别用BBB评分和斜板试验观察大鼠运动功能的恢复情况。免疫组化方法和Western blot方法检测三组大鼠脊髓损伤部位Fas和Caspase1的表达。结果术后7d和14d,GM1治疗组BBB评分和斜板试验明显优于脊髓损伤组(<0.05)。假手术组大鼠脊髓仅有少量Fas和Caspase1阳性表达和阳性细胞,脊髓损伤后1d时则明显增多,7 d开始减弱,但仍多于假手术组(<0.05)。与相同时间点的SCI组比较,GM1组大鼠脊髓损伤部位的Fas和Caspase1阳性表达和阳性细胞明显减少(<0.05)。结论 GM1治疗急性脊髓损伤可能与下调Fas和Caspase1蛋白的表达相关。Objective To study the effect of monosialoganglioside(GM1)on the expressions of Fas and Caspase1 in spinal cord of acute injury rat.Methods 72 SD healthy adult rats were randomly divided into the sham group(24),the spinal cord injury group(24),GM1 group(24)at different time points(1d,7d,14d)after injury.The spinal cord injury(SCI)model was induced with Nystrom's way.At the 1d,7d and 14 d after SCI,the recovery of motor function after SCI in rats was assessed with Basso-Beattie-Bresnahan(BBB)scale and tilt board test.The expressions of Fas and Caspase1 were observed by immunochemistry and western blot methods.Results At the 7d and 14 d after SCI,BBB scores and tilt board test were better in GM1 group than in SCI group.A few positive cells and lower expression levels of Fas and Caspase1 were found in sham group,but the cell number and expression levels increased obviously at 1d and 7 days in SCI group,but decreased in GM1 group than in SCI group.Conclusion The therapeutic effect of GM1 on acute spinal cord injury might be related to the downregulation of the expressions of Fas and Caspase1.
