摘　　要：目的:探讨丹参多酚酸盐对阿托伐他汀钙在心肌缺血大鼠体内的药动学影响。方法 :选取雄性SD大鼠给予腹腔注射垂体后叶素建立心肌缺血损伤模型,将其随机分为两组:单用阿托伐他汀钙组(30 mg·kg-1)与联合用药组(阿托伐他汀钙30 mg·kg-1+丹参多酚酸盐200 mg·kg-1),每组5只。分别于给药前及给药后0.083、0.17、0.25、0.5、1、2、4、8、12、24、36 h从尾静脉采血1 m L,并分离血浆0.5 m L,采用LC-MS/MS测定阿托伐他汀钙的血药浓度,比较两组间药代动力学参数,利用DAS3.0软件进行统计学处理。结果 :合用丹参多酚酸盐后阿托伐他汀钙在心肌缺血大鼠体内药代动力学参数AUC0-t、AUC0-∞增加显著(P<0.05),其它药代动力学参数无显著性差异(P>0.05)。结论:在心肌缺血的病理状态下,丹参多酚酸盐能显著提高阿托伐他汀钙的生物利用度,但对其体内的消除无显著影响。Objective： To study the effect of salvianolate on the pharmacokinetics of atorvastatin calcium in rats withmyocardial ischemia.Methods： The rat models of myocardial ischemia were established by injecting pituitrin into the peritonealcavity of SD rats. Parallel control method was applied in this study. Ten model rats were evenly randomized into two groups：atorvastatin calcium （30 mg·kg-1） group and drug combination group （atorvastatin calcium 30 mg·kg-1 plus salvianolate 200 mg·kg-1）.Blood samples （1 mL） were collected from the caudal vein at 0.083, 0.17, 0.25, 0.5, 1, 2, 4, 8, 12, 24, 36 h after drug administration.LC-MS/MS was used to determine the plasma concentrations of atorvastatin calcium. The pharmacokinetic parameters of the two groupswere compared, and statistical analysis was performed by DAS 3.0 software. Results： When combined with salvianolate, the AUC0-tand AUC0-∞ of atorvastatin calcium were increased obviously （P 〈 0.05）. The other pharmacokinetic parameters had no significantchanges （P 〉 0.05）. Conclusion： In the state of myocardial ischemia, salvianolate could apparently increase the bioavailability ofatorvastatin calcium, while it almost had no effect on drug elimination process.
