摘 要:目的观察铁皮石斛多糖(PDC)对糖尿病大鼠视网膜及血清中炎症因子表达水平的影响,探讨PDC在早期糖尿病视网膜病变中的抗炎作用及机制。方法采用链脲佐菌素(STZ)腹腔注射的方法诱导SD大鼠糖尿病模型。模型建立成功6周后,分别用低剂量PDC[100 mg/(kg·d)]、中剂量PDC[200 mg/(kg·d)]及高剂量PDC[300 mg/(kg·d)]灌胃治疗。8周后,2%戊巴比妥钠(40 mg/kg)麻醉大鼠,取所需标本。采用蛋白印迹法(Westen Blot)检测大鼠视网膜内白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、血管内皮生长因子(VEGF)的表达水平,酶联免疫吸附试验(ELISA)检测大鼠血清IL-6、TNF-α的含量,免疫组化观察大鼠视网膜内核转录因子κB(NF-κB)的表达水平。结果与正常组比较,糖尿病组大鼠视网膜内IL-6、TNF-α、VEGF的蛋白表达增加;血清IL-6、TNF-α表达增加,差异均具有统计学意义(P<0.05)。与糖尿病组比较,PDC低、中、高各剂量组大鼠视网膜内IL-6、TNF-α的蛋白表达明显降低,血清IL-6、TNF-α的表达降低,差异均具有统计学意义(P<0.05)。PDC各剂量组之间各指标两两比较差异均无统计学意义(P>0.05)。免疫组化切片观察,正常大鼠视网膜未见明显NF-κB阳性染色,糖尿病组大鼠视网膜内NF-κB表达高于正常组,与糖尿病组比较,PDC各剂量组NF-κB表达降低。结论 PDC可以降低糖尿病视网膜病变大鼠体内炎症因子(IL-6、TNF-α)的表达,进而抑制VEGF表达上调。其作用机制可能与NF-κB通路有关。OBJECTIVE To investigate the influence of polysaccharides of dendrobium candidum(PDC) on overexpression of inflammatory factors in diabetic retinopathy model of rats and its anti-inflammatory effects in ear-ly-stage diabetic retinopathy and further to explore the possible mechanism. METHODS The diabetic rat model was established by intraperitoneal injection of streptozotocin. After 6 weeks, they were administrated with DPC in low dosage [100 mg/(kg·d)], middle dosage [200 mg/(kg·d)] and high dosage [300 mg/(kg·d)]. After 8 weeks, these animals were anesthetized for specimen collection. Then the protein expression of Interleuldn-6(IL-6), tumor necrosis factor-α(TNF-α) and vascular endothelial growth factor(VEGF) were detected by Western blot; the expression of IL-6, TNF-α in the serum of rats were determined by Enzyme-Linked Immunosorbent Assay(ELISA);the expression of NF-κB p65 in retina was determined by immunohistochemistry. RESULTS Compared with normal group, the protein level of IL-6, TNF-α and VEGF as well as serum IL-6 and TNF-α in diabetic group were increased(P〈0.05) while these indexes all decreased when compared with diabetic group(P〈0.05). But each marker showed no differences between three PDC groups(P〉 0.05). There was no positive staining of NF-κB in normal group, and the expression of NF-κB in three DPC groups was significantly lower than counterpart in diabetic group whose digit was higher than normal group.CONCLUSIONS PDC could reduce the expression of inflammatory cytokines(IL-6, TNF-α)in diabetic retinopathy rats, and inhibit the VEGF expression. The mechanism might be associated with NF-κB pathway.
