摘  要:目的研究甲氨蝶呤(MTX)对中药复方银屑灵2号主要成分芍药苷药代动力学参数的影响,评价联合用药的合理性。方法将12只SD大鼠随机分为银屑灵2号组、联合用药组(MTX+银屑灵2号),单次口服给药后在不同时间点眼眶静脉采血,固相萃取(SPE)处理,采用HPLC测定血浆中芍药苷浓度,观察芍药苷在两组中的药代动力学参数的变化。结果与银屑灵2号组比较,联合用药组芍药苷AUC_(0-t)增加了29.50%,C_(max)增加了35.32%,T_(max)延迟了0.067 h,清除率降低了41.99%。结论 MTX与银屑灵2号联用可增加银屑灵2号中芍药苷的血药浓度,降低清除率,提高生物利用度。Objective To research the impact of Methotrexate(MTX)on the pharmacokinetics of paeoniflorin,which is the main component of Chinese medicine Yinxieling Recipe 2,and to evaluate the rationality of combination therapy.Methods Twelve Sprague- Dawley rats were divided randomly into Yinxieling Recipe 2 group and combined group(MTX combined with Yinxieling Recipe 2),6 rats in each group. After single- dose oral administration,blood was sampled at different time points, and plasma samples were prepared by solid phase extraction method. Plasma paeoniflorin concentration was determined by HPLC method,and the changes of pharmacokinetics of paeoniflorin in the two groups were observed. Results Compared with Yinxieling Recipe 2 group, the AUC0-tof the combined group increased by 29.50 %,Cmax increased by 35.32 %,Tmax extended for 0.067 hours and the clearance rate decreased by41.99 %. Conclusion The drug concentration of paeoniflorin can be increased,the bioavailability is enhanced,and clearance rate is decreased when Yinxieling Recipe 2 is co- administrated with MTX.
