摘 要:目的优选祛风除痹颗粒剂的提取工艺条件。方法运用高效液相色谱法(HPLC)测定补骨脂素、异补骨脂素含量,流动相甲醇-水(45∶55),检测波长246 nm,流速1.0 m L·min^(-1),柱温30℃。以补骨脂素、异补骨脂素含量及固形物总量的综合评分为指标,采用L_9(3~4)正交试验设计考察加水量、提取时间、提取次数对祛风除痹颗粒水提工艺的影响。结果优选提取工艺条件为加8倍量水提取3次,每次1 h。固形物总量、补骨脂素及异补骨脂素含量分别为39.20,25.62,20.35 mg·m L^(-1)。结论该工艺稳定可行,可为祛风除痹颗粒剂的工业化生产提供试验依据。Objective To optimize the extraction technology of Qufeng Chubi granules. Methods The contents of psoralen and isopsoralen were determined by HPLC. The mobile phase consisted of acetonitrile- water(45:55) with a flow rate of 1.0 mL·min^-1 and the detection wavelength was set at 246 nm. The temperature of column was kept at 30℃. With the contents of psoralen and isopsoralen and the comprehensive scores of total volume of solid matters as observation indexes,L_9(3-4)orthogonal design was applied to test the possible effects of water volume,extracting time and extraction frequency on the water- extraction technology of Qufeng Chubi granules. Results Optimum water- extraction technology conditions were as follows: boiling the herbs with eight- fold water for three times, one hour for each time,and the total volume of solid matters,psoralen content and isopsoralen content being 39.2,25.62 and 20.35 mg·mL^-1,respectively. Conclusion The method is reasonable and stable,which will supply evidence for industrial production of Qufeng Chubi granules.
