依那西普对大鼠肺间质病变及CTGF表达的影响

摘　　要：目的观察依那西普对博来霉素诱导的大鼠肺间质病变及肺组织中肺组织结缔组织生长因子（CTGF）含量的影响。方法雄性sD大鼠54只，随机分为3组，即对照组、模型组、治疗组（依那西普），每组18只。采用博来霉素诱导间质性肺病大鼠模型，治疗组予依那西普干预。观察比较造模后第7、14、28天各组大鼠肺组织病理学、羟脯氨酸及CTGF表达的变化。结果治疗前，模型组及治疗组可见明显肺泡炎症、胶原沉积和纤维化改变。治疗组治疗后各时期肺泡炎和肺纤维化程度均较模型组减轻。治疗组治疗后各时期羟脯氨酸的表达较模型组减少（P〈0．05）。免疫组织化学分析显示，造模第28天模型组大鼠肺组织中的CTGF表达显著增加，且治疗组较模型组表达显著减少（P〈0．05）。结论依那西普可减轻博来霉素诱导的肺间质病变大鼠的肺纤维化程度，其机制可能与抑制CTGF表达有关。Objective To.explore the effects of etanercept on the pathological changes of rats with interstitial lung disease （ILD） and their levels of connective tissue growth factor （CTGF）. Methods Male Sprague - Dawley （SD） rats were randomly divided into three groups： a control group, a model group and an etanereept treatment group （n = 18 ）. A rat model of interstitial lung disease was established through bleomyein induction. Meanwhile, etanereept were adminis- trated in the treatment group. On Days 7, 14 and 28, the pathological changes of each group were observed, while the levels of hydroxyproline （HYP） and CTGF were detected in lung tissues. Results Before treatment, alveolar inflamma- tion, collagen deposition and fibrosis changes could be clearly observed in the model and treatment group. After treat- ment, the treatment group showed relieved alveolitis and fibrosis than the model group at each time point. Compared with the model group, the level of HYP was reduced in the treatment group at each time point （P 〈 0.05 ）. According to im- munochemical analysis, on Day 28, enhanced expression of CTGF was found in the model group; in contrast, the treat- ment group produced remarkable less amounts of CTGF （ P 〈 0.05 ）. Conclusion Etanercept can relieve the pulmonary fibrosis in ILD rat,which may be related to the inhibition of CTGF expression.
