摘 要:目的探讨低强度超声联合氨基糖苷类药物对产超广谱β-内酰胺酶(ESBLs)大肠杆菌生物膜内细菌活性及生物膜渗透性的影响。方法超声单独以及分别联合庆大霉素或妥布霉素处理ESBLs大肠杆菌生物膜,测定实验前后生物膜内细菌活性、生物膜大体形态、生物膜对氨基糖苷类药物渗透作用的改变情况。结果 (1)经实验处理后,生物膜内细菌活性出现了明显的下降。(2)扫描电镜、共聚焦结果显示超声辐照前后生物膜结构改变明显。(3)低强度超声联合氨基糖苷类药物能明显增强生物膜对环丙沙星的渗透性。结论低强度超声联合氨基糖苷类药物对ESBLs大肠杆菌生物膜有显著的协同增效杀菌作用,且能改变其渗透性。Objective To study the effects of low-intensity ultrasound combined with aminoglycosides on bacterial vitality and penetration of extended-spectrum beta-lactamases(ESBLs)-producing Escherichia coli(E.coli)biofilms.Methods ESBLs E.coli biofilms were established in vitro,and irradiated by ultrasound with or without antibiotics.The morphology and vitality of bacteria were measured before and after the treatments.A ciprofloxacin penetration test was also conducted.Results(1)After treatment,the biofilms vitality experienced a significant decrease.(2)The results of scanning electron microscope and laser scanning confocal microscope showed that the biofilm morphology was changing.(3)The permeability of biofilmst to aminoglycosides was enhancing significantly after treatment.Conclusions Low-intensity ultrasound with aminoglycosides can enhance the permeability of biofilms,and produce different synergistic bactericidal effects.
