摘 要:目的对牦牛心脏配伍和家猪心脏配伍八味沉香散的丁香酚药代动力学进行比较研究。方法采用HPLC法测定大鼠血浆中丁香酚的含量。以3,4-二甲氧基苯乙烯为内标;使用ALLtima HP C18色谱柱(250 mm×4.6 mm,5μm),柱温为35℃;流动相为乙腈和水,按梯度洗脱,流量为1.0 m L/min;检测波长为280 nm。使用软件DAS 2.0对灌胃配伍不同动物心脏八味沉香散的大鼠体内丁香酚平均血药浓度-时间进行隔室模型拟合。结果以二室模型(权重为1/cc)计算血药浓度与实测值契合度优于其他模型。配伍牦牛心脏八味沉香散和配伍家猪心脏八味沉香散中丁香酚在大鼠体内的最大峰质量浓度(质量分数)Cmax分别为16.21 mg/L和12.50 mg/L;达峰时间Tmax分别为21.67 min和19.17 min;分布半衰期T1/2α分别为20.03 min和42.34 min;消除半衰期T1/2β均为69.315 min。结论在两种不同动物心脏配伍的八味沉香散中丁香酚代谢情况不相同。配伍牦牛心脏组方的代谢较好,其最大峰质量浓度更高、体内转运速率更快、吸收进入血液循环总量更多。Objective To compare the clove phenol pharmacokinetic in Baweichenxiangsan with yak's and pig's heart. Methods By HPLC determination of eugenol in rat plasma. 3,4-dimetboxy styrene as internal standard; ALLtima HP C18 column(250 mm × 4. 6 mm, 5 μm), column temperature was 35 ℃ ; mobile phase was acetonitrile and water, according to the gradient, flow was 1.0 mL/min; detection wavelength was 280 nm. Eugenol average plasma concentration-time for compartment model fit by software DAS 2. 0, for garage Baweichenxiangsan with different animal's heart to rats. Results In two-compartment model( weight of 1/ce) to calculate the plasma concentration and the measured value to fit better than other models. Compatibility yak's heart and pig's heart Baweichenxiang Powder eugenol maximum peak concentration in rats( mass fraction) Cmax were 16. 21 mg/L and 12. 50 mg/L; up peak time T was 21.67 rain and 19. 17 min; distribution half-life T1/2β were 20. 03 min and 42. 34 min; elimination half-life T1/2β were 69.315 min. Conclusion In both eugenol cardiac metabolism compatibility of different animals in different Baweichenxiangsan. Metabolic compatibility yak has better heart prescription, the maximum peak concentration of higher quality, faster internal transfer rates, the total amount absorbed into the blood circulation more.
