摘  要:目的研究苏州汉族人群Toll样受体4(TLR4)单核苷酸多态性与2型糖尿病(T2DM)的关系。方法检测100例T2DM患者(A组)和100例健康体检者(B组)的血压和生化指标等,采用PCR-限制性片段长度多态性(PCR-RELP)法、限制性核酸内切酶和琼脂糖电泳对TLR4Asp299和Thr399多态性进行基因分型。结果 A组SBP、DBP、BMI、FBG、LDL-C、TG和HbA1c均高于B组(P<0.01或P<0.05)。A组和B组均为TLR4Asp299和Thr399纯合子,未发现有TLR4Asp299Gly和Thr399Ile多态性。结论苏州汉族人群T2DM的发生可能与TLR4Asp299Gly和Thr399Ile的多态性无关。Objective To investigate the relationship between single nucleotide polymorphism of Toll-like receptor 4(TLR4)and type 2diabetes mellitus(T2DM)in Chinese Han population in Suzhou.Methods The blood pressure and biochemical index were measured in 100 patients with T2DM(group A)and 100 healthy people(group B).Then the genotypes of TLR4Asp299 and Thr399were detected by PCR-RELP,restriction endonuclease and agarose electrophoresis.Results SBP,DBP,BMI,FBG,LDL-C,TG and HbA1 cin group A were higher than those in group B(P〈0.01 or P〈0.05).TLR4Asp299 and Thr399homozygote without polymorphism could be found in groups of A and B.Conclusion The development of T2 DM may be not closely related with polymorphism of TLR4Asp299 Gly and Thr399 Ile in Chinese Han population in Suzhou.
